Abstract
Tanaman sambiloto (Andrographis paniculata Ness) adalah salah satu tanaman obat yang banyak digunakan untuk mengobati berbagai penyakit di Indonesia. Senyawa kimia bioaktif utama, andrografolid, telah dilaporkan memiliki berbagai aktivitas farmakologi, salah satunya sebagai antidiabetes. Tujuan penelitian ini adalah untuk mengetahui profil bioavailabilitas isolat andrografolid pada sampel plasma kelinci menggunakan antikoagulan EDTA. Hewan yang digunakan pada penelitian ini adalah kelinci ras New Zealand berjenis kelamin jantan dengan berat 3 kg. Hewan uji diberi dosis 8,75 mg melalui jalur pemberian peroral. Pengambilan darah dilakukan pada interval waktu: 0; ½; 1; 1; 1 ½ ; 2; 4; dan 6 jam. Pengujian kadar isolat andrografolid pada plasma kelinci dilakukan menggunakan KCKT dengan fase gerak metanol (65%:35%). Hasil pengamatan diolah menggunakan PK Solver Microsoft Excel 2019 pada model satu kompartemen ekstravaskular untuk mendapatkan profil bioavailabilitas. Berdasarkan hasil penelitian ini, didaptkan ratarata kadar puncak (Cpmaks) isolat andrografolid adalah 4,5006 µg/mL, waktu puncak (Tmaks) adalah pada 1,4103 jam, rata-rata waktu paruh (T½) adalah 1,662 jam, bersihan (klirens) adalah 0,6905 µg/mL/jam, volume adalah 1,6538 µg/mL, serta luas area di bawah kurva konsentrasi terhadap waktu (AUC0-∞) atau bioavailabilitas adalah 19,6216 µg/mL.jam. ---- The sambiloto plant (Andrographis paniculata Ness) is one of the medicinal plants widely used to treat various diseases in Indonesia. The main bioactive chemical compound, andrografolid, has been reported to have various pharmacological activities, one of which is antidiabetic. Andrografolid reduce HbA1c in patients with type 2 diabetes. The purpose of this study was to determine the bioavailability profile of andrografolid isolate in rabbit plasma using EDTA anticoagulant. The animals used in this study was male New Zealand rabbits weighing 3 kg. The animals were administered singel dose of 8.75 mg andrografolid isolate peroral. Blood collection was carried out at time intervals: 0; ½; 1; 1; 1 ½; 2; 4; and 6 hours. The level of andrografolid in rabbit plasma was carried out using HPLC. The isocratic mode of mobile phase consist of methanol and water (65%: 35%). The pharmacokinetics parameter ware analyzed PK Solver Microsoft Excel 2019 on an extravascular onecompartment model to obtain a bioavailability profile. Based on the results of this study, the average peak level (Cpmaks) of andrografolid isolate was 4.5006 µg/mL, the peak time (Tmax) was at 1.4103 hours, the average half-life (T½) was 1, 662 hours, clearance (clearance) was 0.6905 µg/mL/hour, volume was 1.6538 µg/mL, and the area under the concentration to time curve (AUC0-∞) or bioavailability was 19.6216 µg/mL. hour
