Abstract

------- Transdermal drug delivery presents several advantages over oral administration, including bypassing first-pass metabolism, avoiding gastric degradation, and reducing gastrointestinal side effects. However, the outermost layer of the skin, known as the stratum corneum, acts as a primary barrier that limits passive drug permeation, thereby restricting this delivery route to drugs with specific physicochemical properties. Dissolving Microneedles (DMN) are polymer-based drug delivery systems that physically penetratethe stratum corneum, facilitating the efficient delivery of various active substances—including macromolecules and hydrophilic molecules—without causing pain. This literature review aims to investigate the impact of physicochemical properties of active substances and the geometric design of DMN needles on the efficiency of transdermal drug delivery. The review findings indicate that physicochemical parameters such as molecular weight, log P value, and solubility are significant determinants of a substance’s suitability for use within the DMN system. Additionally, needle geometry—including shape, height, diameter, and tip angle—has a demonstrable effect on the mechanical strength of DMN, its skin penetration capability, and its dissolution profile. The conical needle shape has demonstrated superior performance regarding skin insertion ratio and dissolution rate compared to alternative shapes. A comprehensive understanding of the relationship between physicochemical properties of active substances and the geometric design parameters of DMN is essential for optimizing formulations to enhance transdermal therapeutic efficacy.
