Abstract
Kurkumin memiliki aktivitas farmakologi luas namun dibatasi oleh kelarutan rendah dalam medium aqueous, menyebabkan bioavailabilitas oral dan penetrasi kulit yang rendah. Fitosom merupakan sistem penghantaran obat yang menggabungkan senyawa aktif dengan fosfolipid melalui ikatan hidrogen, meningkatkan kelarutan dan permeabilitas membran. Penelitian ini bertujuan mempelajari karakteristik fitosom kurkumin-soya lesitin yang terbentuk dan mendapatkan variasi formula fitosom kurkumin-soya lesitin yang paling optimal. Fitosom kurkumin dibuat dengan lima variasi rasio kurkumin:soya lesitin dengan penambahan kolesterol 10% menggunakan metode thin film hydration. Metode analisis kurkumin divalidasi berdasarkan kriteria ICH. Karakterisasi meliputi ukuran partikel dan indeks polidispersitas (DLS), potensial zeta (LDV), morfologi (TEM), analisis gugus fungsi (FTIR), serta efisiensi enkapsulasi dan kapasitas pemuatan. Formula 1 (rasio 1:1) menunjukkan karakteristik optimal dengan ukuran partikel 270,8 ± 27,02 nm, PDI 0,4784 ± 0,0934, potensial zeta -40,09 ± 2,886 mV, morfologi spherical dengan struktur berlapis, efisiensi enkapsulasi 98,89 ± 0,02%, dan kapasitas pemuatan 47,09 ± 0,01%. Fitosom kurkumin Formula 1 dengan rasio kurkumin:soya lesitin 1:1 merupakan formula paling optimal, menunjukkan karakteristik fisikokimia terbaik dengan agregasi minimal dan stabilitas koloid yang sangat baik. ------ Curcumin exhibits diverse pharmacological activities but is limited by extremely low aqueous solubility, resulting in poor oral bioavailability and skin penetration. Phytosomes are drug delivery systems that combine active compounds with phospholipids through hydrogen bonding, enhancing apparent solubility and membrane permeability. This research aimed to study the characteristics of curcumin-soy lecithin phytosomes formed and obtain the most optimal variation of curcumin-soy lecithin phytosomal formula. Curcumin phytosomes were prepared with five variations of curcumin:soy lecithin ratio with 10% w/w cholesterol addition using thin film hydration method. Curcumin analytical method was validated according to ICH criteria. Characterization included particle size and polydispersity index (DLS), zeta potential (LDV), morphology (TEM), functional group analysis (FTIR), entrapment efficiency and loading capacity determination. Formula 1 (1:1 ratio) showed optimal characteristics with particle size 270.8 ± 27.02 nm, PDI 0.4784 ± 0.0934, zeta potential -40.09 ± 2.886 mV, spherical morphology with layered structures, entrapment efficiency 98.89 ± 0.02%, and loading capacity 47.09 ± 0.01%. Curcumin phytosome Formula 1 with 1:1 curcumin:soy lecithin ratio represents the most optimal formula, showing the best physicochemical characteristics with minimal agglomeration and excellent colloidal stability.
