Abstract
Stres oksidatif berperan dalam kerusakan neuron dan perkembangan penyakit neurodegeneratif. Andrografolid memiliki aktivitas antioksidan yang berpotensi mempertahankan fungsi sel dalam kondisi oksidatif. Penelitian ini bertujuan mengevaluasi pengaruh andrografolid terhadap viabilitas sel neuroblastoma manusia SH-SY5Y yang dipapar hidrogen peroksida (H₂O₂). Penelitian eksperimental in vitro dilakukan dengan memaparkan sel pada H₂O₂ 100 µM selama 24 jam dan memberikan andrografolid pada konsentrasi 0,5; 1; 1,5; 2; 2,5; 3; dan 3,5 µM. Nasetilsistein 2 mM digunakan sebagai kontrol positif. Viabilitas diukur menggunakan WST-8 pada 450 nm dengan panjang gelombang referensi 620 nm. Data berasal dari tiga replikasi teknis dalam satu eksperimen dan disajikan sebagai rerata ± simpangan baku. Viabilitas kontrol sel, H₂O₂, dan NAC + H₂O₂ masing-masing sebesar 100,00 ± 6,86%; 106,51 ± 5,91%; dan 107,69 ± 2,35%. Andrografolid menghasilkan viabilitas 101,01–120,12%, dengan nilai tertinggi pada 1 µM sebesar 120,12 ± 5,17%, tanpa pola dosis–respons monoton. Paparan H₂O₂ belum membentuk model cedera sel berdasarkan parameter WST-8. Andrografolid 0,5–3,5 µM tidak menunjukkan sitotoksisitas, tetapi efek neuroprotektifnya belum dapat dibuktikan pada kondisi penelitian ini. ---- Oxidative stress contributes to neuronal damage and neurodegenerative disease progression. Andrographolide possesses antioxidant activity that may preserve cellular function under oxidative conditions. This study evaluated the effect of andrographolide on the viability of human SH-SY5Y neuroblastoma cells exposed to hydrogen peroxide (H₂O₂). In this in vitro experiment, cells were exposed to 100 µM H₂O₂ for 24 hours and treated with andrographolide at concentrations of 0.5, 1, 1.5, 2, 2.5, 3, and 3.5 µM. N-acetylcysteine at 2 mM served as the positive control. Viability was measured using the WST-8 assay at 450 nm with a 620 nm reference wavelength. Three technical replicates were analyzed and reported as mean ± standard deviation. Viability in the cell control, H₂O₂, and NAC + H₂O₂ groups was 100.00 ± 6.86%, 106.51 ± 5.91%, and 107.69 ± 2.35%, respectively. Andrographolide produced viability values of 101.01–120.12%, with the highest value at 1 µM (120.12 ± 5.17%), without a monotonic dose–response pattern. H₂O₂ exposure did not establish cellular injury based on WST-8. Andrographolide at 0.5– 3.5 µM was not cytotoxic, but its neuroprotective effect could not be demonstrated under these conditions.
