Abstract
Bunga telang (Clitoria ternatea L.), kayu secang (Caesalpinia sappan L.), dan adas manis (Pimpinella anisum L.) merupakan tanaman yang umum dikonsumsi masyarakat dalam bentuk seduhan herbal. Ketiga bahan ini sering dicampur dengan takaran berdasarkan kebiasaan tanpa data ilmiah mengenai kandungan senyawa aktif dan manfaat kesehatannya. Penelitian ini bertujuan menetapkan kadar flavonoid total dan aktivitas antioksidan dari tiga formula kombinasi (F1=1,0:0,8:0,2; F2=1,2:0,6:0,2; F3=1,4:0,4:0,2) serta membandingkannya dengan masing-masing simplisia tunggal. Penetapan kadar flavonoid total dilakukan dengan spektrofotometri sinar tampak menggunakan pereaksi AlCl₃ pada panjang gelombang 433 nm. Aktivitas antioksidan diuji dengan metode DPPH pada panjang gelombang 517 nm dan dinyatakan sebagai nilai IC₅₀. Hasil penelitian menunjukkan Formula 3 menghasilkan kadar flavonoid total tertinggi (2,1535 mg QE/g) dan aktivitas antioksidan terkuat (IC₅₀ 10,53 ppm) di antara ketiga formula, bahkan sedikit lebih baik dibandingkan Bunga Telang tunggal. Seluruh sampel tergolong antioksidan sangat kuat dengan IC₅₀ kurang dari 50 ppm. Uji statistik menunjukkan perbedaan signifikan antar sampel serta korelasi negatif yang kuat antara kadar flavonoid dan aktivitas antioksidan. Simpulan penelitian ini adalah Formula 3 dengan perbandingan 1,4:0,4:0,2 merupakan formula optimal yang berpotensi dikembangkan sebagai minuman herbal berbasis bahan alam. ------ Butterfly pea flower (Clitoria ternatea L.), sappan wood (Caesalpinia sappan L.), and anise (Pimpinella anisum L.) are plants commonly consumed as herbal infusions. These three ingredients are often mixed in proportions based on habit without scientific data regarding their active compound content and health benefits. This study aimed to determine total flavonoid content and antioxidant activity of three combination formulas (F1=1.0:0.8:0.2; F2=1.2:0.6:0.2; F3=1.4:0.4:0.2) and compare them with each single simplicia. Total flavonoid content was determined using visible spectrophotometry with AlCl₃ reagent at 433 nm. Antioxidant activity was tested using DPPH method at 517 nm and expressed as IC₅₀ value. The results showed that Formula 3 produced the highest total flavonoid content (2.1535 mg QE/g) and the strongest antioxidant activity (IC₅₀ 10.53 ppm) among the three formulas, even slightly better than Butterfly Pea Flower alone. All samples exhibited very strong antioxidant activity with IC₅₀ values below 50 ppm. Statistical analysis showed significant differences among samples and a strong negative correlation between flavonoid content and antioxidant activity. It can be concluded that Formula 3 with a ratio of 1.4:0.4:0.2 is the optimal formula with potential to be developed as a natural-based herbal beverage.
