Abstract

Tanaman tin (Ficus carica L.) merupakan tanaman langka di Indonesia yang memiliki banyak manfaat dan digunakan sebagai obat berbagai penyakit seperti diabetes, antibakteri, disentri, kolesterol, dan asam urat. Namun, budidaya tanaman tin di Indonesia banyak dijumpai kendala, sehingga diperlukan teknik perbanyakan secara in vitro melalui teknik kultur jaringan sebagai alternatif budidaya. Penelitian ini bertujuan untuk menentukan konsentrasi optimum dalam proses multiplikasi tunas tanaman tin serta mengetahui karakteristik kandungan metabolit sekunder tanaman tin hasil multiplikasi. Multiplikasi planlet tanaman tin dibagi 8 kelompok dengan variasi konsentrasi zat pengatur tumbuh BAP (0,5 ppm; 1 ppm; 1,5 ppm; 2 ppm), NAA (0,1 ppm; 0,2 ppm; 0,3 ppm; 0,4 ppm) kemudian dilakukan penapisan fitokimia pada eksplan hasil multiplikasi, semua proses tersebut dilakukan secara triplo. Pengamatan dilakukan selama 4 minggu, meliputi warna eksplan secara kualitatif, serta jumlah tunas, daun, akar, dan panjang eksplan secara kuantitatif, kemudian hasil data dianalisis menggunakan ANOVA. Hasil pengamatan menunjukkan adanya perbedaan bermakna pada kelompok uji BAP 1,5 ppm rata- rata jumlah tunas 9,67 dan jumlah daun 11,67; BAP 2 ppm rata-rata jumlah tunas 7,67 dan jumlah daun 13,67; NAA 0,3 ppm rata-rata jumlah akar 13,67dan NAA 0,4 ppm rata-rata jumlah akar 14,00. Konsentrasi optimum terhadap multiplikasi tunas tanaman tin terdapat pada konsentrasi BAP 1,5 ppm. Hasil penapisan fitokimia menunjukkan adanya seyawa flavonoid, alkaloid, tanin, fenolat, steroid dan triterpenoid, kuinon serta monoterpen dan seskuiterpen.;---Fig ara (Ficus carica L.) is a rare plant in Indonesia that has many benefits and it’s used as a medicine for various diseases such as diabetes, antibacterial, dysentery, cholesterol, and gout. However, cultivation of fig ara in Indonesia gets many obstacles. Therefore, in vitro propagation techniques are needed through tissue culture techniques as an alternative to cultivation. This study aims to determine the optimum concentration in the multiplication process of fig ara plants shoots and determine the characteristics of secondary metabolite content of fig ara plants resulting by multiplication. Multiplication of plant planlets divided into 8 groups variations in the concentration of plant growth regulators BAP (0,5 ppm; 1 ppm; 1,5 ppm; 2 ppm), NAA (0,1 ppm; 0,2 ppm; 0,3 ppm; 0,4 ppm) then, phytochemical screening was carried out on explants resulting from multiplication, all processes were carried out in triplo. Observations were made for 4 weeks, including qualitative explant colors, as well as the number of shoots, leaves, roots, and explant length quantitatively, then the results of the data were analyzed using ANOVA. The observations showed a significant difference in the BAP test group 1.5 ppm with an average of 9,67 shoots and 11,67 leaves; BAP 2 ppm with an average of shoots 7,67 and 13,67 of leaves; NAA 0,3 ppm with an average of 13,67 roots and NAA 0,4 ppm with an average of 14,00 roots. The optimum concentration of multiplication of tin plant shoots is found at a BAP concentration of 1.5 ppm. Phytochemical screening results show the presence of flavonoids, alkaloids, tannins, phenolics, steroids and triterpenoids, quinones and monoterpenes and sesquiterpenes.
