Abstract

Biosurfaktan merupakan senyawa aktif yang disintesis oleh mikroorganisme dan memiliki gugus hidrofilik dan hidrofobik, sehingga dapat menurunkan tegangan permukaan. Penelitian ini bertujuan untuk menentukan aktivitas dan karakterisasi biosurfaktan dari bakteri Staphylococcus epidermidis dan Exigobacterium profundunm. Metode yang digunakan pada penelitian ini yaitu penapisan isolat bakteri dengan uji hemolisis dan uji oil spreading, kemudian produksi biosurfaktan dengan pengocokan konstan selama 7 hari, uji aktifitas biosurfaktan menggunakan uji emulsifikasi dengan variasi waktu pengamatan hari ke 0, 1, 3, 5, dan hari ke 7, kromatografi lapis tipis dan uji antibakteri. Pada uji hemolisis kedua bakteri menunjukkan hasil negatif dan hasil uji oil spreading menunjukkan kedua bakteri positif dengan penyebaran lapisan minyak membentuk zona bening, dari uji emulsifikasi diperoleh hasil persentase emulsifikasi Staphylococcus epidermis berurutan yaitu 44,83%, 44,83%, 48,28%, 46,43%, dan 51,72%, persentase emulsifikasi Exigobacterium profundunm berurutan yaitu 44,83%, 48,28%, 48,28%, 40%, dan 43,75%. Pada Hasil KLT menunjukkan adanya bercak berpendar dibawah sinar UV 366 nm dan terbentuk noda merah dengan penampak bercak ninhidrin menunjukkan biosurfaktan termasuk golongan lipopeptida dan biosurfaktan dari kedua bakteri memiliki aktivitas antibakteri. Bakteri Staphylococcus epidermidis dan Exigobacterium profundunm dapat menghasilkan biosurfaktan yang termasuk golongan biosurfaktan lipopeptida dan memiliki aktivitas antibakteri terhadap bakteri Escherichia coli dan Staphylococcus aureus.;---Biosurfactants are active compounds that are synthesized by microorganisms and have hydrophilic and hydrophobic groups, so they can reduce surface tension. This study aims to determine the activity and characterization of biosurfactants from the bacteria Staphylococcus epidermidis and Exigobacterium profundunm. The method used in this research is screening of bacterial isolates by Hemolysis test and Oil spreading test, then biosurfactant production with constant shaking for 7 days, biosurfactant activity test using emulsification test with a variations of observation time days 0, 1, 3, 5, and 7th day , layer chromatography thin and Antibacterial test. the hemolysis test showed negative results on both bacteria and the results of the oil spreading test showed the two positive bacteria with the spread of the oil layer forming a clear zone, from the emulsification test obtained the results of sequential Staphylococcus epidermis emulsification results were 44.83%, 44.83%, 48.28%, 46.43%, and 51.72%, the percentage of Exigobacterium Profundunm emulsification were 44.83%, 48.28%, 48.28%, 40%, and 43.75%, respectively. The TLC results showed that there were glowing spots under UV light 366 nm and a red stain formed with the appearance of ninhydrin spots showed biosurfactants including lipopeptide and biosurfactant groups from both bacteria had antibacterial activity. Staphylococcus epidermidis and Exigobacterium profundunm can produce biosurfactants which belong to the lipopeptide biosurfactant group and have antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria.
