Abstract
Penyakit Alzheimer merupakan salah satu bentuk demensia yang ditandai dengan degenerasi progresif sel saraf, terutama di hipokampus dan korteks serebri. Stres oksidatif akibat induksi aluminium klorida (AlCl₃) dapat memicu kerusakan neuron. Andrografolid, senyawa aktif dari Andrographis paniculata, diketahui memiliki aktivitas antioksidan dan antiinflamasi yang berpotensi sebagai neuroprotektor. Penelitian ini bertujuan untuk mengevaluasi efek neuroprotektif isolat andrografolid terhadap kerusakan sel piramidal hipokampus dan korteks serebri pada tikus putih jantan yang diinduksi AlCl₃. Penelitian eksperimental in vivo ini menggunakan 30 ekor tikus putih jantan galur Wistar yang dibagi menjadi 6 kelompok (normal, kontrol positif, kontrol negatif, serta tiga kelompok dosis andrografolid 5; 7,5; dan 10 mg/kgBB). Induksi AlCl₃ dilakukan secara oral selama 7 hari, kemudian perlakuan isolat andrografolid diberikan selama 28 hari. Analisis histologi dilakukan dengan pewarnaan Hematoksilin-Eosin, dan kerusakan sel piramidal diamati berdasarkan degenerasi inti (piknosis, karioreksis, dan kariolisis). Data dianalisis menggunakan uji ANOVA dan Kruskal–Wallis dengan tingkat signifikansi p<0,05. Induksi AlCl₃ meningkatkan kerusakan sel piramidal secara signifikan dibanding kelompok normal. Pemberian donepezil (kontrol positif) menurunkan tingkat kerusakan. Isolat andrografolid menunjukkan efek neuroprotektif dengan variasi sesuai dosis. Hasil uji statistik menunjukkan dosis 7,5 mg/kgBB memberikan hasil terbaik, ditandai dengan tingkat kerusakan sel yang paling rendah dan tidak berbeda signifikan dengan kontrol positif maupun kelompok normal. ------ Alzheimer’s disease is a form of dementia characterized by progressive degeneration of nerve cells, particularly in the hippocampus and cerebral cortex. Oxidative stress induced by aluminum chloride (AlCl₃) can trigger neuronal damage. Andrographolide, the active compound of Andrographis paniculata, is known to possess antioxidant and anti-inflammatory properties with potential neuroprotective effects. This study aimed to evaluate the neuroprotective effects of andrographolide isolate on pyramidal cell damage in the hippocampus and cerebral cortex of male Wistar rats induced with AlCl₃. This in vivo experimental research used 30 male Wistar rats, divided into six groups (normal, positive control, negative control, and three groups receiving andrographolide at doses of 5, 7.5, and 10 mg/kg BW). AlCl₃ induction was administered orally for 7 days, followed by andrographolide treatment for 28 days. Histological analysis was conducted using Hematoxylin-Eosin staining, and pyramidal cell damage was assessed based on nuclear degeneration (pyknosis, karyorrhexis, and karyolysis). Data were analyzed using ANOVA and Kruskal–Wallis tests with a significance level of p<0.05. AlCl₃ induction significantly increased pyramidal cell damage compared with the normal group. Administration of donepezil (positive control) reduced the extent of damage. Andrographolide isolates exhibited neuroprotective effects with dose-dependent variation. Statistical analysis showed that the 7.5 mg/kg BW dose provided the best results, indicated by the lowest level of cell damage, and was not significantly different from the positive control or the normal group.
