Abstract

Fungi endofit merupakan mikroorganisme yang hidup di jaringan tanaman tanpa menimbulkan gejala penyakit serta berpotensi menghasilkan metabolit bioaktif serupa dengan inangnya. Penelitian ini bertujuan mengisolasi dan mengidentifikasi fungi endofit dari tanaman sambiloto (Andrographis paniculata) melalui optimasi metode isolasi untuk memperoleh isolat murni yang representatif. Optimasi dilakukan terhadap tiga parameter utama, yaitu waktu perendaman pada proses sterilisasi permukaan jaringan, perbandingan efektivitas media pertumbuhan, serta konsentrasi antibiotik kloramfenikol untuk menekan kontaminasi bakteri. Hasil menunjukkan bahwa kombinasi perendaman waktu yang lebih lama, penggunaan media MEA, dan penambahan antibiotik 70 ppm merupakan kondisi paling efektif dalam menghasilkan koloni endofit tanpa kontaminasi. Dari lima isolat murni yang diperoleh, isolat MBP2U dipilih karena pertumbuhan lebih stabil. Secara morfologi, MBP2U membentuk koloni krem kecoklatan hingga keabuan bertekstur beludru, dengan hifa bercabang dan konidia bulat–oval. Analisis molekuler berdasarkan sekuens ITS diperoleh panjang fragmen 548 bp menunjukkan kesamaan 100% dengan Aspergillus versicolor pada BLAST NCBI, serta hubungan kekerabatan kuat dengan spesies tersebut pada analisis filogenetik. Penelitian ini memberikan informasi mengenai prosedur isolasi yang sesuai dan identifikasi fungi endofit potensial dari sambiloto untuk pengembangan metabolit bioaktif secara berkelanjutan. ------- Endophytic fungi are microorganisms that live within plant tissues without causing disease symptoms and have the potential to produce bioactive metabolites similar to those of their host. This study aimed to isolate and identify endophytic fungi from Andrographis paniculata through optimization of the isolation method to obtain representative pure isolates. The optimization involved three main parameters: immersion time during surface sterilization, comparison of growth media effectiveness, and chloramphenicol antibiotic concentration to suppress bacterial contamination. The results showed that a longer immersion time, the use of MEA medium, and the addition of 70 ppm chloramphenicol were the most effective conditions for producing uncontaminated endophytic colonies. Of the five pure isolates obtained, isolate MBP2U was selected due to its stable growth. Morphologically, MBP2U formed cream to grayish-brown, velvety colonies with branched hyphae and round–oval conidia. Molecular analysis based on an ITS sequence of 548 bp showed 100% similarity with Aspergillus versicolor according to NCBI BLAST, and a strong monophyletic relationship with this species in phylogenetic analysis. This study provides information on appropriate isolation procedures and the identification of potential endophytic fungi from sambiloto for the sustainable development of bioactive metabolites.
