Abstract

Andrografolid merupakan senyawa bioaktif utama dari sambiloto (Andrographis paniculata Nees) yang digunakan sebagai marker dalam analisis fitokimia. Penelitian ini bertujuan mengevaluasi efektivitas metode Solid Phase Extraction (SPE) sebagai tahap preparasi andrografolid dari ekstrak sambiloto dan isolatnya menggunakan Kromatografi Cair Kinerja Tinggi (KCKT) dan Spektrofotometri UV-Visibel. Tahapan SPE meliputi pengkondisian, pemuatan sampel, pencucian, dan elusi dengan variasi pelarut aqua pro injection, buffer asetat, serta metanol 40%. Kondisi optimum diperoleh dengan pelarut pengkondisian metanol murni, pencucian aqua pro injection 1 mL, dan elusi metanol 1 mL. Analisis KCKT menunjukkan konsentrasi andrografolid tertinggi pada ekstrak sebesar 50,442 µg/mL, sedangkan spektrofotometri UV-Visibel sebesar 106,224 µg/mL pada kondisi yang sama. Sedangkan pada isolat, analisis KCKT menunjukkan konsentrasi andrografolid sebesar 39,227 µg/mL, sedangkan spektrofotometri UV-Visibel sebesar 61,105 µg/mL pada kondisi yang sama. Uji t-test menunjukkan perbedaan signifikan (p < 0,05) antara kedua metode pada sampel ekstrak, namun tidak signifikan pada isolat. Hasil ini mengindikasikan bahwa KCKT memiliki selektivitas lebih tinggi terhadap matriks kompleks, sementara spektrofotometri memberikan hasil sebanding pada isolat. Dengan demikian, SPE efektif digunakan untuk preparasi dan pemurnian andrografolid dari ekstrak sambiloto dengan sistem aqua pro injection. ------- Andrographolide is the main bioactive compound of Andrographis paniculata Nees, which is used as a marker in phytochemical analysis. This study aims to evaluate the effectiveness of the Solid Phase Extraction (SPE) method as a preparative step for andrographolide from sambiloto extract and its isolate using High Performance Liquid Chromatography (HPLC) and UV-Visible Spectrophotometry. The SPE stages include conditioning, sample loading, washing, and elution with variations of aqua pro injection solvent, acetate buffer, and 40% methanol. Optimal conditions were obtained with pure methanol as the conditioning solvent, 1 mL of aqua pro injection for washing, and 1 mL of methanol for elution. HPLC analysis showed the highest concentration of andrographolide in the extract at 50.442 µg/mL, while UVVisible spectrophotometry showed 106.224 µg/mL under the same conditions. Meanwhile, in the isolate, KCKT analysis showed an andrographolide concentration of 39.227 µg/mL, while UV-Visible spectrophotometry showed 61.105 µg/mL under the same conditions. The t-test showed a significant difference (p < 0.05) between the two methods in the extract sample, but not in the isolate. These results indicate that KCKT has higher selectivity toward complex matrices, while spectrophotometry provides comparable results in isolates. Thus, SPE is effective for the preparation and purification of andrographolide from sambiloto extract using the aqua pro injection system.
