Abstract
Alfa gamma sodium mangostin merupakan senyawa bioaktif golongan xanton yang telah melalui proses penggaraman dan banyak terkandung dalam kulit buah manggis (Garcinia mangostana L.), serta memiliki berbagai aktivitas terapi. Dengan banyaknya penggunaan kulit manggis dalam pengobatan maka perlu dilakukan uji keamanan senyawa alfa gamma sodium mangostin melalui uji toksisitas akut oral. Tujuan dari penelitan adalah untuk menentuan nilai LD50 dan tingkat keamanan senyawa alfa gamma sodium mangostin dengan menggunakan metode OECD 425 Up and Down Procedure, serta pengaruhnya terhadap histopatologi organ lambung, hati dan ginjal. Data yang diperoleh dianalisis menggunakan program AOT425StatPgm untuk menentukan nilai LD50. Pada penelitian ini mencit diberi isolat alfa gamma sodium mangostin dengan dosis 5000 kg/kg BB dan diamati gejala toksisitas serta perubahan bobot badan mencit selama 14 hari. Hasil penelitian menunjukkan bahwa tidak teramati adanya gejala toksisitas maupun kematian hewan uji. Pada pengamatan makroskopik tidak teramati tanda-tanda toksisitas pada organ lambung, hati, dan ginjal akibat pemberian isolat alfa gamma sodium mangostin. Namun, pada pengamatan mikroskopik terjadi perubahan patologik pada jaringan lambung berupa erosi, namun pada jaringan hati dan ginjal tidak ditemukan adanya tanda-tanda toksisitas. Nilai LD50 senyawa alfa gamma sodium mangostin yang diperoleh yaitu >5000 mg/kg BB dengan kategori senyawa uji praktis tidak toksik.;---Alfa sodium gamma mangostin is a group of xanton bioactive compounds who has gone through the process of salting and contained in the rind of the mangosteen (Garcinia mangostana L.), and has various therapeutic activities. With the many uses of mangosteen rind in treatment, it is necessary to test the safety of alpha gamma sodium mangostin compounds through oral acute toxicity tests. The aim of this study to determine the LD50 value and the safety level of the alpha gamma sodium mangostin compound using the OECD 425 Up and Down Procedure method, and its effect againts the histopathology of the stomach, liver and kidney organs. The obtained data were analyzed using the AOT425StatPgm program to determine LD50 values. In this study, the mice were given alpha gamma sodium mangostin isolates at 5000 kg / kg BB of dose and observed the symptoms of toxicity and changes in body weight of mice for 14 days. The results showed that no symptoms of toxicity or death of animals test. In macroscopic observations, no signs of toxicity in the stomach, liver, and kidney organs due to the administration of alpha gamma sodium mangostin isolates. However, in microscopic observations there are pathological changes in gastric tissue in the form of erosion, but in the liver and kidney tissue there are no signs of toxicity. The LD50 values of alpha gamma sodium mangostin compounds obtained was > 5000 mg / kg BB which is the category of test compounds was practically non-toxic.
