Abstract

Radikal bebas berperan dalam patogenesis penyakit degeneratif sehingga diperlukan sumber antioksidan alami yang aman dan berkelanjutan. Berdasarkan mekanismenya, antioksidan menetralkan radikal bebas terutama melalui donasi elektron atau atom hidrogen, sehingga memutus reaksi berantai oksidatif. Penelitian ini menilai aktivitas penangkap radikal bebas dari ampas ekstrak kunyit (Curcuma domestica Val.) sebagai bentuk pemanfaatan limbah herbal. Metode yang digunakan adalah uji DPPH dengan pembacaan pada 517 nm; nilai IC₅₀ ditentukan melalui regresi linier, dan vitamin C digunakan sebagai pembanding. Skrining fitokimia pada ampas menunjukkan flavonoid, alkaloid, saponin, dan triterpenoid positif; sedangkan polifenol dan tanin negatif. Hasil uji menunjukkan nilai IC₅₀ ampas sebesar 264,452 µg/mL (kategori sangat lemah), sedangkan vitamin C 25,529 µg/mL (sangat kuat). Temuan ini mengindikasikan kandungan fenolik efektif pada ampas relatif rendah akibat proses ekstraksi, sehingga kemampuan penangkapan radikal terbatas. Disimpulkan bahwa ampas ekstrak kunyit masih memiliki aktivitas antioksidan namun tidak memadai sebagai sumber tunggal; optimalisasi proses atau kombinasi dengan bahan kaya fenolik disarankan untuk peningkatan potensi. ----- Free radicals contribute to the pathogenesis of degenerative diseases, necessitating safe and sustainable sources of natural antioxidants. Based on their mechanism, antioxidants neutralize free radicals primarily by donating electrons or hydrogen atoms, thereby terminating oxidative chain reactions. This study evaluated the freeradical-scavenging activity of turmeric extract residue (Curcuma domestica Val.) as a means of utilizing herbal waste. The DPPH assay was employed with readings at 517 nm; IC₅₀ values were determined by linear regression, and vitamin C was used as a comparator. Phytochemical screening of the residue showed positive results for flavonoids, alkaloids, saponins, and triterpenoids, while polyphenols and tannins were negative. The assay yielded an IC₅₀ of 264.452 µg/mL for the residue (very weak category) and 25.529 µg/mL for vitamin C (very strong). These findings indicate that the effective phenolic content in the residue is relatively low due to the extraction process, limiting its radical-scavenging capacity. It is concluded that turmeric extract residue exhibits antioxidant activity but is insufficient as a standalone source; process optimization or combination with phenolic-rich materials is recommended to enhance its potential.
