Abstract
Penyalahgunaan narkotika dan psikotropika, khususnya amfetamin dan turunannya, masih menjadi masalah sosial dan tantangan kesehatan masyarakat yang signifikan di berbagai negara, termasuk Indonesia. Berdasarkan laporan Badan Narkotika Nasional (BNN), prevalensi penyalahgunaan narkotika di Indonesia mencapai 1,73% dari populasi usia produktif. Kondisi tersebut menunjukkan pentingnya pengembangan metode deteksi yang cepat, akurat, dan spesifik untuk mendukung upaya pencegahan serta penanggulangan penyalahgunaan zat tersebut. Penelitian ini bertujuan untuk menentukan afinitas pengikatan fragmen antibodi scFv6H4 terhadap amfetamin menggunakan pendekatan penambatan molekul, serta mengevaluasi spesifisitasnya terhadap narkotika lainnya seperti metamfetamin, kokain, morfin, THC, dan benzodiazepin. Hasil analisis penambatan molekul menunjukkan bahwa interaksi antara scFv6H4 dengan amfetamin memiliki nilai energi ikatan bebas sebesar -7,19 kkal/mol. Sebagai pembanding, nilai energi ikatan bebas ligan lain diperoleh hasil masing-masing, yaitu metamfetamin -7,25 kkal/mol, morfin -6,23, kokain -9,15, benzodiazepin -6,51, dan ganja -8,69. Nilai energi paling negatif ditunjukkan oleh kokain, sedangkan interaksi antara scFv6H4 dengan amfetamin masih berada pada tingkat yang cukup stabil. Temuan ini mengindikasikan bahwa meskipun scFv6H4 memiliki kemampuan berinteraksi dengan amfetamin, tingkat spesifisitasnya masih dipengaruhi oleh afinitas terhadap ligan lain, khususnya kokain. Oleh karena itu, diperlukan optimasi lebih lanjut sebelum scFv6H4 dapat dikembangkan sebagai kandidat utama dalam sistem deteksi berbasis lateral flow assay (LFA) untuk amfetamin. ------ The abuse of narcotics and psychotropic drugs, particularly amphetamines and their derivatives, remains a significant social problem and public health challenge in many countries, including Indonesia. According to a report by the National Narcotics Agency (BNN), the prevalence of drug abuse in Indonesia reaches 1.73% of the productive age population. This condition highlights the importance of developing fast, accurate, and specific detection methods to support efforts to prevent and combat substance abuse. This study aims to determine the binding affinity of the scFv6H4 antibody fragment to amphetamine using a molecular docking approach, as well as to evaluate its specificity to other narcotics such as methamphetamine, cocaine, morphine, THC, and benzodiazepines. The results of the molecular docking analysis showed that the interaction between scFv6H4 and amphetamine had a free binding energy value of -7.19 kcal/mol. For comparison, the free energy values of other ligands were obtained as follows: methamphetamine -7.25 kcal/mol, morphine - 6.23, cocaine -9.15, benzodiazepine -6.51, and marijuana -8.69. The most negative energy value was shown by cocaine, while the interaction between scFv6H4 and amphetamine remained at a fairly stable level. These findings indicate that although scFv6H4 has the ability to interact with amphetamine, its specificity is still influenced by its affinity for other ligands, particularly cocaine. Therefore, further optimization is needed before scFv6H4 can be developed as a leading candidate in a lateral flow assay (LFA)-based detection system for amphetamine.
