Abstract
Tanaman tin (Ficus Carica L.) memiliki banyak manfaat sebagai obat berbagai penyakit. Namun, budidaya tanaman tin di Indonesia banyak dijumpai kendala, sehingga diperlukan teknik perbanyakan dengan kultur jaringan. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi konsentrasi BAP (Benzyl Amino Purin) dan NAA (Naphthalena Acetid Acid) terbaik untuk pertumbuhan tunas tanaman tin dan mengetahui kandungan metabolit sekundernya. Eksplan tanaman tin dibagi menjadi 9 kelompok, kelompok kontrol MS0, kelompok uji zat pengatur tumbuh BAP 0,5 mg/L+NAA (0,1;0,2;0,3;0,4 mg/L) dan BAP1 mg/L+NAA (0,1;0,2;0,3;0,4 mg/L). Pengamatan eksplan dilakukan selama 4 minggu secara kualitatif meliputi warna eksplan dan secara kuantitatif meliputi panjang eksplan, jumlah tunas, jumlah daun dan jumlah akar. Eksplan tanaman tin kemudian diekstraksi dengan etanol 96% dengan metode maserasi. Pada ekstrak dilakukan penapisan fitokimia. Data pengamatan dianalisis menggunakan ANOVA. Hasil penelitian pada minggu ke 4, panjang eksplan terbaik 3,16cm (BAP 1 mg/L + BAP 0,1 mg/L), jumlah Tunas terbaik 10,66 (BAP 1 mg/L + NAA 0,1 mg /L), jumlah daun 8,66 (BAP 1 mg/L + NAA 0,1 mg/L) dan jumlah akar 2,33 (BAP 1 + NAA 0,2 mg/L). Konsentrasi optimum terhadap multiplikasi tanaman tin yaitu dengan penambahan BAP 1 +NAA 0,1 mg/L yang memiliki perbedaan bermakna dalam setiap pengamatan kuantitatif dibandingkan dengan kelompok kontrol. Hasil penapisan fitokimia ekstrak etanol eksplan tanaman tin menunjukkan adanya senyawa golongan alkaloid, flavonoid, fenolat, tanin, monoterpen dan seskuiterpen dan saponin;---Tin plants (Ficus Carica L.) have many benefits as medicines for various diseases. However, there are many obstacles encountered in tin cultivation in Indonesia, so that the propagation technique is needed by tissue culture. This study aims to determine the effect of the combination of the best concentration of BAP (Benzyl Amino Purin) and NAA (Naphthalena Acetid Acid) for the growth of tin plant buds and knowing the content of secondary metabolites. tin plant explants were divided into 9 groups, MS0 control group, growth regulator test group BAP 0.5 mg / L + NAA (0.1; 0.2; 0.3; 0.4 mg / L) and BAP1 mg / L + NAA (0.1; 0.2; 0.3; 0.4 mg / L). Explanatory observations carried out for 4 weeks qualitatively included explant color and quantitatively included explants length, number of shoots, number of leaves and number of roots. tin plant explants were then extracted with 96% ethanol by maceration method. Phytochemical screening was carried out on the extract. Observation data were analyzed using ANOVA. The results of the study at week 4, the best explants length was 3.16cm (BAP 1 mg / L + BAP 0.1 mg / L), the best number of shoots was 10.66 (BAP 1 mg / L + NAA 0.1 mg / L) , the number of leaves was 8.66 (BAP 1 mg / L + NAA 0.1 mg / L) and the number of roots was 2.33 (BAP 1 + NAA 0.2 mg / L). The optimum concentration of tin plant multiplication was by adding BAP 1 + NAA 0.1 mg / L which had a significant difference in each quantitative observation compared to the control group. Phytochemical screening results of tin plant ethanol extract showed the presence of alkaloid compounds, flavonoids, phenolics, tannins, monoterpenes and sesquiterpenes and saponins.
