Abstract
Brazilin merupakan senyawa aktif Biopharmaceutics Classification System kelas IV dengan kelarutan rendah dan bioavailabilitas terbatas membuat efek farmakologinya kurang maksimal. Penelitian ini bertujuan mengevaluasi peningkatan kelarutan, profil disolusi, dan stabilitas fisik brazilin dalam bentuk dispersi padat amorf (DPA) menggunakan polimer hidrofilik PVP K-30 melalui metode solvent evaporation. Formula dibuat dengan rasio brazilin:PVP K-30 sebesar 1:1, 1:3, dan 1:5. Evaluasi dilakukan melalui uji kelarutan menggunakan magnetic stirer dan disolusi menggunakan dissolusion tester serta dianalisis menggunakan spektrofotometri UV-Vis serta uji stabilitas fisik dengan X-Ray Diffraction (PXRD) setelah penyimpanan 30 hari pada kelembapan 20% dan 90%. DPA rasio 1:1 meningkatkan kelarutan hingga dua kali lipat, sedangkan rasio 1:3 dan 1:5 meningkat 1–1,5 kali lipat. Profil disolusi menunjukkan efek “spring and parachute” dengan pelepasan tertinggi pada rasio 1:1 pada menit 15 (60%), dibandingkan 1:3 pada menit 15 (27%) dan 1:5 pada menit 2 (36%). Stabilitas terbaik juga ditunjukkan oleh formula 1:3 yang tetap mempertahankan bentuk amorf. Formula 1:1 dan 1:5 membentuk kristal pada hari ke-7. Berdasarkan penelitian ini, dapat disimpulkan bahwa DPA rasio 1:1 merupakan formula paling efektif dalam meningkatkan kelarutan, disolusi, dan stabilitas brazilin. ------ Brazilin is an active compound classified under Class IV of the Biopharmaceutics Classification System, with poor water solubility and limited bioavailability, which reduces its pharmacological effectiveness. This study aimed to evaluate the enhancement of solubility, dissolution profile, and physical stability of brazilin in the form of amorphous solid dispersion (ASD) using the hydrophilic polymer PVP K-30 via the solvent evaporation method. Formulations were prepared with brazilin:PVP K-30 ratios of 1:1, 1:3, and 1:5. Evaluations included solubility testing using a magnetic stirrer, dissolution testing using a dissolution tester, both analyzed with UV-Vis spectrophotometry, and physical stability testing via X-Ray Diffraction (XRD) after 30 days of storage at 20% and 90% relative humidity. The 1:1 ASD formulation increased solubility up to twofold, while the 1:3 and 1:5 ratios showed 1 to 1.5-fold improvements. Dissolution profiles indicated a "spring and parachute" effect, with the highest release observed in the 1:1 ratio at 15 minutes (60%), compared to 1:3 at 15 minutes (27%) and 1:5 at 2 minutes (36%). The best physical stability was shown by the 1:3 formulation, which maintained its amorphous form, while the 1:1 and 1:5 formulations exhibited recrystallization on day 7. Based on these results, the 1:1 ASD is the most effective formulation for enhancing the solubility, dissolution, and stability of brazilin.
