Abstract

Obesitas merupakan masalah kesehatan global yang ditandai oleh penumpukan lemak tubuh secara berlebih dan dapat memicu berbagai penyakit degeneratif. Daun cincau hitam (Mesona palustris Blume) mengandung flavonoid, saponin, tanin, dan serat yang berpotensi menurunkan berat badan. Penelitian ini bertujuan untuk mengevaluasi pengaruh serbuk Komponen Pembentuk Gel (KPG) daun cincau hitam terhadap berat badan dan indeks Lee pada mencit obesitas (Mus musculus), serta menentukan dosis efektifnya. Sebanyak 30 ekor mencit betina dibagi menjadi enam kelompok: kontrol normal, kontrol negatif, kontrol positif (Orlistat Xenical®), serta tiga kelompok perlakuan dengan dosis KPG 100, 200, dan 300 mg/kgBB. Induksi obesitas dilakukan melalui pakan tinggi lemak selama 14 hari, dilanjutkan dengan pemberian KPG selama 14 hari. Parameter yang diamati adalah perubahan berat badan, panjang tubuh, dan indeks Lee. Data dianalisis menggunakan uji normalitas, homogenitas, ANOVA satu arah, dan post hoc LSD. Hasil menunjukkan penurunan berat badan (p = 0,003) dan indeks Lee (p = 0,027) yang signifikan antar kelompok. Dosis 300 mg/kgBB menunjukkan penurunan indeks Lee tertinggi. Indeks Lee dinilai lebih representatif karena mempertimbangkan rasio berat badan terhadap panjang tubuh. Kesimpulannya, serbuk KPG daun cincau hitam berpotensi menurunkan obesitas, terutama pada dosis 300 mg/kgBB, dengan efek lebih jelas pada indeks Lee. ----- Obesity is a global health problem characterized by excessive fat accumulation and can trigger various degenerative diseases. Black cincau leaves (Mesona palustris Blume) contain flavonoids, saponins, tannins, and fiber, which have the potential to reduce body weight. This study aimed to evaluate the effect of Black Cincau Leaf Gel-Forming Component (GFC) powder on body weight and Lee index in obese mice (Mus musculus), as well as to determine its effective dose. A total of 30 female mice were divided into six groups: normal control, negative control, positive control (Orlistat Xenical®), and three treatment groups receiving GFC powder at doses of 100, 200, and 300 mg/kgBW. Obesity was induced using a high-fat diet for 14 days, followed by GFC powder administration for another 14 days. Observed parameters included changes in body weight, body length, and Lee index. Data were analyzed using normality and homogeneity tests, one-way ANOVA, and post hoc LSD test. The results showed significant reductions in body weight (p = 0.003) and Lee index (p = 0.027) between groups. The 300 mg/kgBW dose showed the greatest decrease in Lee index. The Lee index is considered more representative as it accounts for the ratio of body weight to body length. In conclusion, GFC black grass jelly leaf powder has the potential to reduce obesity, especially at a dose of 300 mg/kgBW, with a more significant effect on the Lee index.
