Abstract

Andrographis paniculata merupakan tanaman herbal yang memiliki beragam aktivitas farmakologis seperti antiinflamasi, antioksidan dan antikanker. Namun, kandungan andrografolid alami pada tanaman cenderung rendah, sehingga diperlukan pendekatan bioteknologi seperti kultur jaringan dengan penambahan zat pengatur tumbuh (ZPT) dan elisitor untuk meningkatkan produksinya. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan ZPT 2,4-D dan kinetin serta elisitor kitosan dalam upaya meningkatkan kadar andrografolid pada kalus yang dihasilkan dari tanaman sambiloto. Dalam penelitian ini, kalus diinisiasi dari eksplan daun sambiloto pada media Murashige and Skoog (MS) dengan tiga variasi konsentrasi ZPT 2,4-D dan kinetin (2,5: 1,5 mg/L, 2 :1mg/L dan 1,5: 0,5 mg/L. Kalus yang terbentuk kemudian diekstraksi menggunakan metanol dan dianalisis secara kuantitatif menggunakan Kromatografi Cair Kinerja Tinggi (KCKT) untuk menentukan perlakuan terbaik sebelum diberi perlakuan elisitor kitosan. Hasil penelitian menunjukkan bahwa kombinasi ZPT dengan konsentrasi 1,5 mg/L : 0,5 mg/L menghasilkan kandungan andrografolid tertinggi yaitu dengan rata-rata kadar 1,921%. Pada perlakuan elisitor kitosan 75 dan 100 ppm dengan waktu inkubasi 7 dan 9 hari, dianalisi kembali menggunakan KCKT dan hasil penelitian menunjukkan konsentrasi elisitor 100 ppm dengan waktu inkubasi 9 hari menghasilkan kadar andrografolid tertinggi yaitu 3,012%. Penambahan elisitor kitosan berpengaruh pada produksi andrografolid, pada konsentrasi 100 ppm hari ke 9 menghasilkan perlakuan paling optimal dalam merangsang peningkatan senyawa andrografolid pada kultur kalus sambiloto. -------- Andrographis paniculata is a medicinal plant with various pharmacological activities, such as anti-inflammatory, antioxidant, and anticancer properties. However, the natural andrographolide content in the plant tends to be low, making it necessary to apply biotechnological approaches such as plant tissue culture with the addition of plant growth regulators (PGRs) and elicitors to enhance its production. This study aimed to investigate the effect of adding the PGRs 2,4-D and kinetin and the elicitor chitosan in increasing andrographolide content in callus derived from A. paniculata. In this research, callus was initiated from A. paniculata leaf explants on Murashige and Skoog (MS) medium with three variations of 2,4-D and kinetin concentrations (2.5:1.5 mg/L, 2:1 mg/L, and 1.5:0.5 mg/L). The formed callus was then extracted using methanol and quantitatively analyzed by High-Performance Liquid Chromatography (HPLC) to determine the best treatment before applying the chitosan elicitor. The results showed that the combination of 1.5 mg/L 2,4-D : 0.5 mg/L kinetin yielded the highest andrographolide content, with an average of 1.921%. In the chitosan elicitor treatments (75 and 100 ppm) with incubation periods of 7 and 9 days, further analysis using HPLC revealed that the 100 ppm chitosan treatment with 9 days of incubation produced the highest andrographolide content, reaching 3.012%. The addition of chitosan elicitor significantly influenced andrographolide production, with the 100 ppm concentration at 9 days being the most optimal treatment to stimulate andrographolide accumulation in A. paniculata callus cultures.
