Abstract

Hiperlipidemia merupakan keadaan patologis akibat kelainan metabolisme lemak yang ditandai dengan meningkatnya kadar kolesterol. Keadaan hiperlipidemia menyebabkan peningkatan radikal bebas sehingga dapat menurunkan antioksidan di dalam tubuh yang utamanya Superoxide Dismutase (SOD). Isolat andrografolid mengandung flavonoid diketahui memiliki efek sebagai antioksidan dari penelitian ini adalah untuk mengetahui pengaruh pemberian isolat andrografolid terhadap kadar SOD pada kelompok tikus hiperlipidemia. Penelitian ini menggunakan hewan model tikus umur 8-10 minggu dengan BB 200-350 gram. Pembuatan keadaan hiperlipidemia dilakukan dengan cara pemberian pakan kaya lemak dan penambahan Propiltiourasil (PTU) selama 28 hari. Tikus dibagi menjadi enam kelompok, yaitu kelompok normal, skit, simvastatin, dosis 1 sebesar 2 mg/kgBB, dosis 2 sebesar 4 mg/kgBB dan dosis 3 sebesar 6 mg/kgBB selama 28 hari. Analisis data kuantitatif menggunakan one way Analysis of Variance (ANOVA). Hasil penelitian menunjukkan pemberian isolat andrografolid dapat meningkatkan kadar SOD pada hati tikus secara signifikan (p 0,05) antar kelompok perlakuan Dosis 3 sebanyak 6 mg/kg BB merupakan dosis terbaik untuk meningkatkan kadar SOD sebesar 59,61% terhadap kelompok hiperlipidemia dapat memperbaiki kerusakan organ hepar tikus hiperlipidemia. Kesimpulan dari penelitian ini adalah isolat andrografolid dapat meningkatkan kadar SOD sebagai antioksidan di dalam tubuh. --- Hyperlipidemia is a pathological condition resulting from abnormal lipid metabolism characterized by elevated cholesterol levels. The condition of hyperlipidemia leads to an increase in free radicals, which can reduce antioxidants in the body, primarily Superoxide Dismutase (SOD). Isolated andrographolide contains flavonoids known to have antioxidant effects. The purpose of this study is to determine the effect of isolated andrographolide administration on SOD levels in a group of hyperlipidemic rats. This study used an animal model of rats aged 8-10 weeks with a body weight of 200-350 grams. The induction of hyperlipidemia was done by providing a high-fat diet and adding Propylthiouracil (PTU) for 28 days. The rats were divided into six groups: normal, control, simvastatin, dose 1 at 2 mg/kg BW, dose 2 at 4 mg/kg BW, and dose 3 at 6 mg/kg BW for 28 days. Quantitative data analysis was performed using one-way Analysis of Variance (ANOVA). The research results showed that the administration of isolated andrographolide could significantly increase SOD levels in the rat liver (p < 0.05) among treatment groups. Dose 3 at 6 mg/kg BW was the most effective dose, increasing SOD levels by 59.61% in the hyperlipidemia group, which could improve liver organ damage in hyperlipidemic rats. The conclusion of this study is that isolated andrographolide can increase SOD levels as an antioxidant in the body.
