Abstract
Rumput mutiara (Oldenlandiae corymbosa L.) dikenal sebagai tanaman herbal yang memiliki potensi aktivitas antioksidan dan telah dimanfaatkan dalam pengobatan tradisional. Penelitian ini bertujuan mengisolasi serta mengidentifikasi senyawa antioksidan dari fraksi n-heksan ekstrak etanol herba rumput mutiara. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol, dilanjutkan dengan partisi cair-cair menggunakan n-heksan, etil asetat, dan air. Fraksi n-heksan dianalisis menggunakan Kromatografi Lapis Tipis (KLT) dan kromatografi kolom gravitasi untuk memisahkan senyawa aktif. Uji aktivitas antioksidan dengan semprotan DPPH (2,2-difenil-1-pikrildrazil) menunjukkan bercak kuning pada latar ungu, menandakan adanya aktivitas antioksidan. Isolat murni diperoleh melalui KLT preparatif dan dikonfirmasi dengan KLT dua dimensi. Identifikasi lebih lanjut menggunakan uap amonia, pereaksi AlCl₃, serta spektrofotometri UV-Vis dengan pereaksi geser mengindikasikan bahwa isolat diduga termasuk golongan fenol dengan sistem konjugasi aromatik. Hasil ini menunjukkan fraksi n-heksan herba rumput mutiara berpotensi sebagai sumber senyawa fenolik antioksidan alami. ------ Oldenlandiae corymbosa L., commonly known as rumput mutiara, is a traditional medicinal plant recognized for its potential antioxidant activity. This study aimed to isolate and identify antioxidant compounds from the n-hexane fraction of the ethanolic extract of O. corymbosa herb. Extraction was performed by maceration using ethanol, followed by liquid–liquid partition with n-hexane, ethyl acetate, and water. The n-hexane fraction was analyzed using Thin Layer Chromatography (TLC) and gravity column chromatography to separate active compounds. Antioxidant activity, evaluated by spraying with DPPH (2,2-diphenyl-1- picrylhydrazyl), showed yellow spots against a purple background, indicating antioxidant presence. A pure isolate was obtained through preparative TLC and confirmed by two-dimensional TLC. Further identification using ammonia vapor, AlCl₃ reagent, and UV–Vis spectrophotometry with shift reagents suggested that the isolate belongs to the phenolic group with an aromatic conjugated system. These findings indicate that the n-hexane fraction of O. corymbosa herb has potential as a natural source of phenolic antioxidant compounds.
