Abstract

Daun kumis kucing (Orthosiphon aristatus) mempunyai senyawa antibakteri. Aktivitas antibakteri pada kombucha dapat ditingkatkan dengan menambahkan tanaman yang mengandung senyawa bioaktif. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari kombucha daun kumis kucing terhadap bakteri Bacillius subtilis, Escherichia coli, Pseudomonas aeruginosa, dan Staphylococcus aureus. Pengujian ini menggunakan metode difusi cakram dengan tiga variasi konsentrasi serbuk simplisia, yaitu 2%, 3%, dan 4%. Amoksisilin digunakan sebagai kontrol positif dan infusa daun kumis kucing sebagai pembanding standar. Hasil penelitian menunjukan bahwa kombucha daun kumis kucing memiliki aktivitas antibakteri spektrum luas. Konsentrasi simplisia 4% menunjukkan zona hambat terbesar terhadap Staphylococcus aureus 15,63 ± 0,1258 mm, sedangkan konsentrasi 2% menunjukkan zona hambat terkecil terhadap Pseudomonas aeruginosa 4,29 ± 0,205 mm. Penghambatan pertumbuhan bakteri lebih baik pada bakteri gram positif daripada gram negatif. Dengan demikian, kombucha daun kumis kucing menunjukan aktivitas antibakteri spektrum luas dan berpotensi sebagai minuman fungsional. ------ Orthosiphon aristatus contains antibacterial compounds. The antibacterial activity of kombucha can be enhanced by incorporating plants that possess bioactive constituents. This study aims to evaluate the antibacterial activity of cat’s whiskers leaf kombucha against Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. The assay was conducted using the disk diffusion method with three variations of simplicia powder concentrations: 2%, 3%, and 4%. Amoxicillin served as the positive control, while the infusion of cat’s whiskers leaves was used as a standard comparator. The findings indicate that cat’s whiskers leaf kombucha exhibits broad-spectrum antibacterial activity. The 4% simplicia concentration produced the largest inhibition zone against Staphylococcus aureus 15.63 ± 0.1258 mm, whereas the 2% concentration yielded the smallest inhibition zone against Pseudomonas aeruginosa 4.29 ± 0.205 mm. The inhibition of bacterial growth was more pronounced against Gram-positive bacteria compared to Gram-negative bacteria. Thus, cat’s whiskers leaf kombucha demonstrates broad-spectrum antibacterial properties and holds potential as a functional beverage.
