Abstract

Perkembangan farmasi veteriner saat ini mendorong para peneliti untuk mengembangkan produk veteriner berbasis bahan alam. Kurkumin dari kunyit (Curcuma longa L.) dan piperin dari lada putih (Piper nigrum L.) dapat digunakan sebagai bahan aktif untuk sediaan emulsi suplemen hewan peliharaan. Penelitian ini bertujuan untuk mengembangkan dan memvalidasi metode Kromatografi Cair Kinerja Tinggi (KCKT) untuk analisis kuantitatif kurkumin dan piperin dalam sediaan emulsi suplemen hewan peliharaan. Sediaan emulsi diformulasikan dengan kombinasi surfaktan tween 80 dan span 80, dengan penambahan propilen glikol, Pulvis Gummi Arabicum (PGA), kalium sorbat, sirupus simpleks, dan akuades. Evaluasi mutu sediaan dilakukan melalui uji organoleptik, volume terpindahkan, cemaran mikroba, cemaran logam berat, berat jenis, pH, stabilitas, dan viskositas, mengacu pada standar BPOM No. 24 Tahun 2023. Metode KCKT menggunakan kolom C-18 dengan fase gerak asetonitril: asam asetat 2% dan deteksi pada panjang gelombang 425 nm untuk kurkumin dan 343 nm untuk piperin. Hasil validasi menunjukkan bahwa metode tersebut memenuhi kriteria linearitas (r ≥ 0,999), akurasi (recovery 98–102%), presisi (RSD < 2%), dan LOD dan LOQ yang memadai. Kadar kurkumin dan piperin dalam sediaan emulsi yang dihasilkan masing-masing sebesar 0,177% dan 12,425%. Sediaan emulsi memenuhi sebagian besar persyaratan mutu BPOM No. 24 Tahun 2023 selama penyimpanan 30 hari, namun parameter pH serta cemaran angka kapang dan khamir masih perlu optimasi lebih lanjut. Meskipun metode analisis telah memenuhi parameter validasi, diperlukan perbaikan untuk analisis simultan kedua senyawa secara optimal. Penelitian ini memberikan kontribusi terhadap pengembangan metode analisis dan formulasi sediaan emulsi suplemen bahan aktif alami untuk hewan peliharaan. ------ The current development of veterinary pharmaceuticals is encouraging researchers to develop veterinary products based on natural ingredients. Curcumin from turmeric (Curcuma longa L.) and piperine from white pepper (Piper nigrum L.) can be used as active ingredients for emulsion preparations of pet supplements. This study aims to develop and validate a High Performance Liquid Chromatography (HPLC) method for quantitative analysis of curcumin and piperine in pet supplement emulsion preparations. The emulsion preparation is formulated with a combination of tween 80 and span 80 surfactants, with the addition of propylene glycol, Pulvis Gummi Arabicum (PGA), potassium sorbate, sirupus simplex, and aquades. Evaluation of the quality of the preparation was carried out through organoleptic tests, displaced volume, microbial contamination, heavy metal contamination, specific gravity, pH, stability, and viscosity, referring to the BPOM No. 24 of 2023 standard. The HPLC method uses a C-18 column with an acetonitrile phase of motion: 2% acetic acid and detection at wavelengths of 425 nm for curcumin and 343 nm for piperine. The validation results showed that the method met the criteria of linearity (r ≥ 0.999), accuracy (recovery 98–102%), precision (RSD < 2%), and adequate LOD and LOQ. The levels of curcumin and piperine in the resulting emulsion preparations were 0.177% and 12.425%, respectively. The emulsion preparation meets most of the quality requirements of BPOM No. 24 of 2023 during 30-day storage, but the pH parameters and mold and yeast contamination still need further optimization. Although the analysis method has met the validation parameters, improvements are needed for the simultaneous analysis of both compounds optimally. This research contributes to the development of methods of analysis and formulation of emulsion preparations of natural active ingredient supplements for pets.
