Abstract

Brazilin merupakan senyawa polifenol yang memiliki aktivitas farmakologi antioksidan dan anti inflamasi. Penelitian ini dilakukan untuk menghasilkan senyawa turunan brazilin melalui reaksi eterifikasi menggunakan benzil klorida. Brazilin di derivatisasi dengan mereaksikannya bersama benzil klorida untuk membentuk turunan eter. Brazilin diisolasi dan dimurnikan kemudian direaksikan dengan benzil klorida menggunakan piperidin sebagai katalis. Hasil reaksi brazilin dipisahkan menggunakan Ekstraksi Cair-Cair (ECC) dan dimurnikan dengan kromatografi kolom. Produk reaksi diidentifikasi menggunakan kromatografi lapis tipis (KLT), spektrofotometer UV-Visibel dan spektrofotometer FTIR. Hasil KLT menunjukkan perubahan posisi noda, mengindikasikan terbentuknya senyawa baru. Analisis spektrofotometer UV-Visibel menunjukkan pergeseran panjang gelombang maksimum dari 445 nm (Brazilin Standar) ke 438 nm (Derivat Brazilin). Spektrum FTIR mengkonfirmasi perubahan gugus fungsi pada senyawa hasil modifikasi. Uji aktivitas antioksidan menggunakan metode DPPH menunjukkan penurunan aktivitas pada senyawa hasil modifikasi, dengan nilai IC50 menurun dari 57 μg/mL (brazilin standar) menjadi 70 μg/mL (Derivat Brazilin). Uji kelarutan menunjukan adanya sedikit perubahan. Tetapi untuk uji stabilitas menunjukan hasil yang sama. Derivatisasi brazilin melalui reaksi eterifikasi dapat mempengaruhi aktivitas antioksidan, meskipun stabilitas dan sifat kelarutan tetap terjaga. ----- Brazilin is a polyphenolic compound that has antioxidant and anti-inflammatory pharmacological activities. This research was carried out to produce brazilin derivative compounds through an etherification reaction using benzyl chloride. Brazilin is derivatized by reacting it with benzyl chloride to form an ether derivative. Brazilin was isolated and purified then reacted with benzyl chloride using piperidine as a catalyst. The brazilin reaction product was separated using Liquid-Liquid Extraction (ECC) and purified by column chromatography. Reaction products were identified using thin layer chromatography (TLC), UV-Visible spectrophotometer and FTIR spectrophotometer. The TLC results showed a change in the position of the stain, indicating the formation of a new compound. UV-Visible spectrophotometer analysis shows a shift in the maximum wavelength from 445 nm (Standard Brazilin) to 438 nm (Brazilin Derivative). The FTIR spectrum confirmed changes in functional groups in the modified compounds. The antioxidant activity test using the DPPH method showed a decrease in activity in the modified compound, with the IC50 value decreasing from 57 μg/mL (standard Brazilin) to 70 μg/mL (Brazilin derivative). The solubility test showed a slight change. But the stability test shows the same results. Derivatization of brazilin through etherification reactions can affect antioxidant activity, although stability and solubility properties are maintained.
