Abstract
Kelarutan merupakan salah satu faktor yang mempengaruhi ketersediaan hayati obat. Diperkirakan lebih dari 40% zat aktif baru yang beredar di pasaran baik yang diperoleh dari hasil sintesis maupun bahan alam memiliki kelarutan yang sangat rendah. Obat yang memiliki kelarutan rendah akan mengakibatkan laju disolusinya juga rendah sehingga absorbsi kurang sempurna. Upaya untuk meningkatkan laju disolusi obat salah satunya dengan teknik kokristalisasi. Metode kokristalisasi dapat dilakukan dengan metode slurry conversion dan liquid assisted grinding. Koformer golongan asam karboksilat yang sering digunakan dalam pembentukan kokristal yaitu, asam benzoat, asam suksinat, asam adipat, asam glutarat, asam maleat, dan lain-lain. Tujuan review jurnal ini adalah untuk mengetahui pengaruh pembentukan kokristal dengan koformer golongan asam karboksilat menggunakan metode slurry conversion dan liquid assisted grinding dapat meningkatkan disolusi zat aktif. Hasil review ini menunjukkan bahwa pembentukan kokristal dengan koformer golongan asam karboksilat menggunakan metode kokristalisasi dapat meningkatkan disolusi dari kokristal AMG 517, piperin dan etionamid beberapa kali lipat lebih tinggi dibandingkan dengan obat murninya. ---- Solubility is one of the factors that affect the bioavailability of drugs. It is estimated that more than 40% of new active substances on the market, both those obtained from synthesis and natural materials, have very low solubility. Drugs that have low solubility will result in a low dissolution rate so that absorption is less than perfect. One of the efforts to increase the rate of drug dissolution is the cocrystallization technique. The cocrystallization method can be done using the slurry conversion and liquid assisted grinding methods. The coformers of the carboxylic acid group that are often used in the formation of cocrystal are benzoic acid, succinic acid, adipic acid, glutaric acid, maleic acid, and others. The purpose of this journal review is to determine the effect of cocrystal formation with carboxylic acid coformers using the slurry conversion and liquid assisted grinding methods to increase the dissolution of active substances. The results of this review indicate that the formation of cocrystal with carboxylic acid coformer using the cocrystallization method can increase the dissolution of AMG 517 cocrystal, piperine and ethionamide several times higher compared to the pure drug.
