Abstract

Piperin memiliki banyak efek farmakologis yang bermanfaat untuk kesehatan seperti antimikroba, antidiare, antijamur dan antiradang. Namun, penggunaan klinis piperin masih terbatas. Salah satu penyebabnya informasi mengenai kelarutan piperin pada pustaka yang ada masih terbatas. Penelitian ini dilakukan untuk memperkuat pustaka yang ada serta menentukan metode penentuan kelarutan piperin yang lebih murah, sederhana, cepat dan cukup handal. Metode yang digunakan ialah Saturation Shake-Flask Method dengan memakai kombinasi analisis gravimetri dan spektrofotometri UV-Visibel. Sampel disonikasi selama 10 menit kemudian diaduk menggunakan shaker 400 rpm selama 24 jam. Sampel dibiarkan mengendap selama 24 jam lalu disaring menggunakan kertas saring bebas air kemudian diuapkan dalam oven 105℃ hingga bobot konstan. Filtrat dilarutkan kembali dengan metanol untuk dianalisis menggunakan spektrofotometri UVVisibel. Kurva kalibrasi piperin dibuat dengan mengukur absorbansi sampel pada panjang gelombang 343 nm. Kurva tersebut menunjukkan linearitas yang baik dengan nilai r sebesar 0,9993. Proses validasi juga memiliki presisi yang baik dengan RSD gravimetri 0,78% dan RSD spektrofotometri 0,92%. Hasil kelarutan menunjukkan pelarut asetonitril memiliki kelarutan terbaik sebesar 0,0488 g/mL sedangkan pelarut air sebagai yang terburuk sebesar 0,0002 g/mL. Berdasarkan hasil penelitian, kepolaran kemungkinan mempengaruhi kelarutan sedangkan log P pelarut tidak mempengaruhi kelarutan piperin dan metode ini dapat digunakan sebagai alternatif pengujian kelarutan piperin. --- Piperine has many pharmacological effects that are beneficial to health such as antimicrobial, antidiarrheal, antifungal and anti-inflammatory. However, the clinical use of piperine is still limited. One of the reasons is the limited information regarding piperine solubility in the existing literature. This research was conducted to strengthen the existing literature and determine the method of determining the solubility of piperine which is cheaper, simpler, faster and quite reliable. The method used is the Saturation Shake-Flask Method using a combination of gravimetric analysis and UV-Visible spectrophotometry. The sample was sonicated for 10 minutes and then stirred using a 400 rpm shaker for 24 hours. The sample was allowed to settle for 24 hours and then filtered using water-free filter paper and then evaporated in an oven at 105 to constant weight. The filtrate was redissolved with methanol for analysis using UV-Visible spectrophotometry. The piperine calibration curve was made by measuring the absorbance of the sample at a wavelength of 343 nm. The curve shows good linearity with an r value of 0.9993. The validation process also has good precision with 0.78% gravimetric RSD and 0.92% spectrophotometric RSD. The solubility results showed that acetonitrile had the best solubility of 0.0488 g/mL while water as the worst solvent was 0.0002 g/mL. Based on the research results, the polarity may affect the solubility while the log P solvent does not affect the piperine solubility and this method can be used as an alternative to piperine solubility testing.
