Abstract

Analisis metanol menggunakan spektrofotometri uv-vis terus dikembangkan untuk mendapatkan metode penetapan kadar yang mudah, salah satunya dengan penambahan reagen pemberi warna yaitu Sodium Nitropruside (SNP), namun metode ini belum tervalidasi sehingga belum dapat digunakan secara luas. Tujuan penelitian ini adalah untuk menetapkan validasi metode analisis metanol dengan spektrofotometer uv-vis dengan penambahan reagen pemberi warna SNP. Parameter pengujian meliputi akurasi, presisi, linearitas, selektivitas, batas deteksi (LOD), batas kuantitasi (LOQ) dan rentang. Hasil penelitian menunjukkan parameter selektivitas tidak masuk kedalam kategori memenuhi syarat, sedangkan parameter validasi yang lain yaitu presisi memberikan rentang 1,6-2,4% (nilai RSD 1,991%), uji akurasi memberikan rentang % perolehan kembali 97-100% (nilai % recovery 98,42%), uji linearitas pada rentang konsentrasi 0 – 2% memberikan nilai koefisien korelasi r = 0.998 dengan batas deteksi (LOD) 0,0575% dan batas kuantifikasi (LOQ) 0,1917 %. Berdasarkan hasil penelitian dapat disimpulkan bahwa spektrofotometri uv-vis pada analisis metanol menggunakan reagen SNP tidak tervalidasi atau tidak valid karena tidak terpenuhinya salah satu kriteria dari tujuh parameter validasi. --- Methanol analysis using uv-vis spectrophotometer continues to be developed to obtain an easy assay method, one of which is the addition of a coloring reagent, namely Sodium Nitropruside (SNP), but this method has not been validated so it cannot be used widely. The purpose of this study was to determine the validation of the methanol analysis method with a uv-vis spectrophotometer with the addition of SNP coloring reagent. The test parameters include accuracy, precision, linearity, selectivity, limit of detection (LOD), limit of quantitation (LOQ) and range. The results showed that the selectivity parameter did not fall into the eligible category, while the other validation parameters, namely precision, gave a range of 1.6-2.4% (RSD value 1.991%), the accuracy test gave a range of 97-100% % recovery (value of % recovery 98.42%), linearity test in the concentration range 0 – 2% gave a correlation coefficient value of r = 0.998 with a detection limit (LOD) of 0.0575% and a quantification limit (LOQ) of 0.1917%. Based on the results of the study, it can be concluded that uv-vis spectrophotometry in methanol analysis using SNP reagents was not validated or invalid because one of the seven validation parameters did not meet the criteria.
