Abstract

Sediaan tablet kombinasi kurkumin, piperin dan mangostin diformulasikan untuk tujuan penggunaan meningkatkan daya tahan tubuh. Informasi mengenai toksisitas dari sediaan ini diperlukan untuk menilai keamanan penggunaannya. Tujuan penelitian ini adalah untuk menentukan nilai LDso dan kategori toksisitas dari sediaan tablet kombinasi kurkumin, piperg dan mangostin serta pengaruhnya terhadap histopatologi organ lambung, hati, ginjal, limpa, dan paru-paru. Metode uji toksisitas akut oral yang digunakan adalah OECD 425 Up and Down Procedure. Data yang diperoleh dianalisis menggunakan program AOT425StatPgm untuk menentukan nilai LDso. Pada penelitian ini tikus diberi bahan uji dengan dosis 2000 mg/kgBB. Pengamatan tanda-tanda toksisitas dilakukan pada jangka pendek (48 jam) dan jangka panjang (14 hari). Hasil penelitian menunjukkan bahwa nilai LDso dari sediaan tablet dari formulasi kombinasi kurkumin, piperin dan mangostin adalah > 2000 mg/kgBB, termasuk dalam kategori toksik ringan. Hasil pengamatan histopatologi menunjukkan bahwa kombinasi kurkumin, piperin dan mangostin dalam sediaan tablet tidak berpengaruh terhadap organ hati, limpa dan paru-paru. Namun, teramati adanya kerusakan parah pada lambung yang ditandai adanya ulkus serta ginjal ditandai dengan adanya atrofi pada glomerulus dan nekrosis pada tubulus. --- Combination tablets of curcumin, piperine and mangostin are formulated for use to increase endurance. Information regarding the toxicity of this preparation is required to assess its safety for use. The purpose of this study was to determine the LDye value and the toxicity category of the combined tablet preparation of curcumin, piperine and mangostin and the treffect on the histopathology of the stomach, liver, kidney, spleen, and lungs. The oral acute toxicity test method used wan OECD 425 Up And Down Procedure. The data obtained were analyzed using the AOT425StatPgm program to determine the LDss value. In this study, rats were given the test material at a dose of 2000 mg/kgBW. Taxicity signs were performed in the short term (48 hours) and long term (14 days). The results showed that the LDss value of tablet preparations from the combination formulation of curcumin piperine and mangostin was 2000 mg/kgBW, included in the mild toxic category Hispathological observations showed that the combination of curcumin, piperine and mangostin in tablet preparations had no effect on the liver, spleen and lungs However, it was observed that there was levere damage to the stomach characterized by ulcers and the kidneys were characterized by atrophy of the glomerulus and necrosis of the tubules.
