Abstract
Penelitian ini bertujuan untuk menentukan kadar fenol total pada ekstrak daun sukun (Artocarpus altilis (parkinson) Fosberg), ekstrak bunga rosella (Hibiscus Sabdariffa L.), serta kombinasi ( ekstrak sukun: ekstrak rosella) 1:1, 1:2, dan 2:1. Penentuan kadar fenol dilakukan secara kualitatif dengan kromatografi lapis tipis (KLT) dan secara kuantitatif dengan Follin-Ciocalteu menggunakan metode spektrofotometri UV-Vis. Metode ekstraksi yang digunakan adalah metode dekoktasi dengan pelarut air. Pada uji KLT, ekstrak air daun sukun dan bunga rosella menghasilkan noda berwarna hitam pada plat KLT setelah disemprot dengan FeCl3 10%, sehingga esktrak teridentifikasi adanya senyawa fenol. Secara kuantitatif, uji dilakukan dengan menggunakan spektrofotometri UV-Vis pada Panjang gelombang 765nm. Hasilnya dinyatakan dalam gram Ekuivalen asam galat (GAE) per 100 gram sampel. Kadar fenol total yang diperoleh adalah 0,546 gram GAE/100gram untuk ekstrak daun sukun, 0,823 gram GAE/100gram untuk ekstrak bunga rosella, 0,798 gram GAE/100gram untuk perbandingan dari ekstrak sukun:ekstrak bunga rosella 1:1, 0,953 gram GAE/100gram untuk perbandingan 1:2 dan 0,800 gram GAE/100gram untuk perbandingan 2:1. Dimana hasil yang paling tinggi terdapat pada kombinasi ekstrak daun sukun dan ekstrak bunga rosella dengan perbandingan 1:2, hasil tersebut mengindikasikan bahwa adanya potensi sinergis yang signifikan antara daun sukun dan bunga rosella dalam meningkatkan kandungan fenol total. ---- This study aims to determine the total phenol content in breadfruit leaf extract (Artocarpus altilis (Parkinson) Fosberg), rosella flower extract (Hibiscus Sabdariffa L.), and their combinations (breadfruit extract: rosella extract) 1:1, 1:2, and 2 :1. Determination of phenol content was carried out qualitatively by thin layer chromatography (TLC) and quantitatively by Follin-Ciocalteu using the UV-Vis spectrophotometry method. The extraction method used is the decoctation method with water solvent. In the TLC test, the water extract of breadfruit leaves and rosella flowers produced black spots on the TLC plate after being sprayed with 10% FeCl3, so the extract was identified as containing phenolic compounds. Quantitatively, testing was carried out using UV-Vis spectrophotometry at a wavelength of 765nm. The results are expressed in grams of gallic acid equivalent (GAE) per 100 grams of sample. The total phenol content obtained was 0.546 grams GAE/100grams for breadfruit leaf extract, 0.823 grams GAE/100grams for rosella flower extract, 0.798 grams GAE/100 grams for breadfruit extract:rosella flower extract 1:1, 0.953 grams GAE/100 grams for a ratio of 1:2 and 0.800 grams GAE/100 grams for a ratio of 2:1. Where the highest results were found in the combination of breadfruit leaf extract and rosella flower extract with a ratio of 1:2, these results indicate a significant synergistic potential between breadfruit leaves and rosella flowers in increasing the total phenol content.
