Abstract

Neurodegenerasi adalah proses penurunan fungsi otak yang terjadi secara perlahan dan progresif disertai dengan gangguan pada memori dan belajar. Isolat andrografolid memiliki potensi meningkatkan fungsi kognitif. Penelitian ini bertujuan untuk mengevaluasi aktivitas neuroprotektif isolat andrografolid menggunakan metode uji Novel Object Recognition (NOR) terhadap tikus jantan galur wistar yang diinduksi aluminium klorida (AlCl₃) sebagai model hewan penyakit neurodegeneratif. Tikus dibagi menjadi 6 kelompok, yaitu: kelompok normal, kelompok negatif yakni kelompok yang hanya diberi induksi tanpa pemberian obat, kelompok positif yakni kelompok yang diberi induksi kemudian diberi obat pembanding citicoline, dan kelompok uji variasi dosis isolat andrografolid 2,5; 5; 7,5 mg/KgBB. Hasil penelitian menunjukkan bahwa pemberian isolat andrografolid secara efektif memperbaiki fungsi memori tikus yang terpapar AlCl₃, seperti yang ditunjukkan oleh peningkatan indeks rekognisi pada tiap kelompok uji. Hasil keseluruhan menunjukan bahwa dosis isolat andrografolid 5mg/KgBB yang paling efektif dalam memperbaiki fungsi kognitif tikus yang diinduksi AlCl₃. Simpulan tersebut diambil karena hasil perbandingan indeks rekognisi pada kelompok uji dosis 5 mg/Kg BB menunjukan perbaikan yang paling tinggi serta hasil statistik T dependent menunjukan perbedaan yang signifikan terhadap indeks rekognisi setelah induksi AlCl₃ dan hasil uji Independent sample T-Test terhadap kelompok negatif juga menunjukan perbedaan signifikan. Uji statistik Independent sample T-Test juga menunjukan kemampuan isolat andrografolid tidak berbeda signifikan dengan kemampuan citicoline sebagai senyawa pembanding. Temuan ini mengindikasikan bahwa isolat andrografolid memiliki potensi sebagai agen neuroprotektif dalam mengatasi defisit memori yang disebabkan oleh paparan AlCl₃. ---- Neurodegeneration is a gradual and progressive decline in brain function accompanied by impairments in memory and learning. Andrographolide isolate has the potential to enhance cognitive function. This study aims to evaluate the neuroprotective activity of andrographolide isolate using the Novel Object Recognition (NOR) test method on male Wistar rats induced with aluminum chloride (AlCl₃) as a model of neurodegenerative disease. The rats were divided into six groups: a normal group, a negative control group that received only the induction without any treatment, a positive control group that was induced and then treated with the comparator drug citicoline, and test groups with varying doses of andrographolide isolate at 2,5, 5, and 7,5 mg/KgBW. The results showed that the administration of andrographolide isolate effectively improved memory function in rats exposed to AlCl₃, as evidenced by an increase in recognition index in each test group. The overall results indicate that the dose of 5 mg/KgBW of andrographolide isolate was the most effective in improving the cognitive function of rats induced with AlCl₃. This conclusion was drawn because the comparison of the recognition index in the 5 mg/KgBW dose test group showed the highest improvement, and the dependent T-test results indicated a significant difference in recognition index after AlCl₃ induction. The Independent Sample T-Test results also showed a significant difference when compared to the negative control group. Furthermore, the Independent Sample T-Test demonstrated that the effectiveness of andrographolide isolate was not significantly different from that of citicoline as a comparator compound. These findings suggest that andrographolide isolate has potential as a neuroprotective agent in addressing memory deficits caused by AlCl₃ exposure.
