Abstract
Kurkumin yang telah terbukti memiliki berbagai efek farmakologis memerlukan studi preformulasi terkait dengan data inkompatibilitas untuk memastikan keamanan dan efektivitas dalam produk obat. Tujuan penelitian ini adalah untuk mengetahui data inkompatibilitas fisikokimia antara zat aktif isolat kurkumin dengan beberapa bahan pengawet yang umum digunakan dalam sediaan farmasi, yaitu nipagin, nipasol, dan natrium benzoat. Metode penelitian ini meliputi pencampuran isolat kurkumin dengan eksipien pengawet (1:1 b/b), diikuti analisis homogenitas, kristalinitas menggunakan XRD, sifat termal menggunakan DSC, struktur kimia dan gugus fungsi dengan FTIR. Hasil penelitian menunjukkan bahwa tidak terdapat pola difraksi baru pada analisis sifat kristalinitas campuran kurkumin dengan ketiga eksipien pengawet. Sementara itu, pada analisis termal menunjukkan adanya pergeseran titik lebur pada termogram DSC untuk campuran kurkumin dengan nipagin dan nipasol. Hasil ini dikonfirmasi oleh spektrum FTIR yang menunjukkan tidak terbentuknya gugus fungsi baru pada campuran kurkumin dengan ketiga eksipien pengawet. Berdasarkan hasil tersebut maka dapat disimpulkan bahwa isolat kurkumin tidak menunjukkan potensi inkompatibilitas yang serius dengan eksipien pengawet nipagin, nipasol, dan natrium benzoat. ---- Curcumin which has been proven to have various pharmacological effects, requires preformulation studies related to incompatibility data to ensure safety and effectiveness in medicinal products. The purpose of this study is to determine physicochemical incompatibility data between the active substance curcumin isolate and several commonly used preservatives in pharmaceutical preparations, namely nipagin, nipasol, and sodium benzoate. The research method includes mixing curcumin isolate with preservative excipients (1:1 w/w), followed by analysis of homogeneity, crystallinity (XRD), thermal properties (DSC), functional groups and chemical structures (FTIR). The results showed that there were no new diffraction patterns in the crystallinity analysis of curcumin mixtures with the three preservative excipients. Meanwhile, thermal analysis showed a shift in melting points on DSC thermograms for curcumin mixtures with nipagin and nipasol. These results were confirmed by FTIR spectra which did not show the formation of new functional groups in the curcumin mixtures with the three preservative excipients. Based on these results, it can be concluded that curcumin isolate does not show serious incompatibility potential with the preservative excipients nipagin, nipasol, and sodium benzoate.
