Abstract

Kulit buah kopi arabika (Coffea arabica L.) merupakan limbah hasil perkebunan yang memiliki potensi sebagai sumber senyawa bioaktif, seperti flavonoid dan fenolik serta antioksidan alami. Penelitian ini bertujuan untuk mengetahui pengaruh metode ekstraksi maserasi dan sonikasi terhadap kandungan total flavonoid, total fenolik, serta aktivitas antioksidan pada ekstrak kulit buah kopi arabika yang berasal dari daerah Rancabali, Bandung. Prosedur penelitian meliputi pengumpulan dan preparasi sampel, pembuatan simplisia, karakterisasi bahan uji, penapisan fitokimia, proses ekstraksi menggunakan metode maserasi selama 3x24 jam dan metode sonikasi selama 1 jam, 2 jam dan 3 jam pada suhu 30°C, serta pengujian aktivitas antioksidan dengan metode DPPH. Analisis kandungan total flavonoid dan total fenolik dilakukan dengan metode spektrofotometri UV-Vis. Hasil penelitian menunjukkan bahwa rendemen ekstrak, kadar total fenolserta totalflavonoid pada ekstrak kulit kopi dengan metode ekstraksi sonikasi lebih tinggi dibandingkan ekstrak hasil metode maserasi. Namun berdasarkan nilai IC₅₀ terhadap DPPH, ekstrak kulit kopi dengan metode maserasi memiliki potensi antioksidan yang lebih baik daripada pada ekstrak hasil sonikasi. Penelitian diharapkan dapat mendukung pemanfaatan limbah kulit buah kopisebagai bahan baku alami pada industri pangan fungsional, kosmetik, dan farmasi. ----- Arabica coffee peel (Coffea arabica L.) is an agricultural by product that has potential as a source of bioactive compounds, such as flavonoids, phenolics, and natural antioxidants. This study aims to determine the effect of maceration and sonication extraction methods on the total flavonoid content, total phenolic content, and antioxidant activity of arabica coffee peel extract sourced from the Rancabali region, Bandung. The research procedures including sample collection and preparation, simplicia production, sample characterization, phytochemical screening, extraction using the maceration method for 3×24 hours and the sonication method for 1 hour, 2 hour, and 3 hour at 30°C, and antioxidant activity testing using the DPPH method. The total flavonoid and total phenolic contents were analyzed using UV-Vis spectrophotometryThe results showed that the extract yield, total phenol and total flavonoid content in coffee skin extract with the sonication extraction method were higher than the extract from the maceration method. However, based on the IC₅₀ value for DPPH, coffee skin extracts extracted using the maceration method have better antioxidant potential than those extracted using sonication.. This research is expected to support the utilization of coffe fruit skin waste as a natural raw material in the functional food, cosmetic, and pharmaceutical industries.
