Abstract
Kanker merupakan penyakit penyebab kematian tertinggi kedua setelah stroke dan hipertensi. Penelitian terkait sintesis bahan alam bertanda radioaktif terbukti mampu mendeteksi dan sekaligus menghancurkan sel kanker dalam tubuh. Berdasarkan informasi tersebut, penelitian ini bertujuan untuk mensintesis senyawa bahan alam kurkumin dengan radioisotop iodium-131 sebagai agen terapi kanker dan menentukan kondisi optimum penandaan hingga memperoleh efisiensi penandaan yang tinggi untuk mencapai tujuan terapi. Dalam percobaannya, senyawa kurkumin telah berhasil ditandai dengan Na-1311 melalui metode kloramin- T. Efisiensi penandaan serta kemurnian radiokimia senyawa ditentukan dengan metode elektroforesis kertas dan kromatografi lapis tipis menggunakan plat ITLC- SG sebagai fase diam dan NaC10,9% sebagai fase gerak. Hasil uji dideteksi dengan TLC-radio scanner (AR2000) dengan kondisi penandaan optimal diperoleh pada penggunaan 0,1 mg/100µl kurkumin, 100 µci Na-1311, 0,625mg/40µl kloramin-T dan natrium metabisulfit, serta waktu inkubasi selama 90 menit pada temperatur 25-30°C. Rata-rata persen kemurnian yang diperoleh sebesar 90,62% ±0,77. Hasil uji lipofilisitas menunjukkan penurunan sifat lipofil dari kurkumin, sedangkan dari hasil elektroforesis menunjukkan senyawa 131-kurkumin bermuatan negatif. Berdasarkan hasil pengujian tersebut, dapat disimpulkan bahwa senyawa bertanda telah memenuhi syarat kemurnian radiokimia sediaan radiofarmaka menurut Farmakope Indonesia dimana syarat minimal kemurnian radiokimia yang ditetapkan adalah >90%. ---- Cancer is the second highest cause of death after stroke and hypertension. Research related to the synthesis of radioactive marked natural materials is proven to detect and simultaneously destroy cancer cells in the body. Based on this information, this study aims to synthesize curcumin natural ingredient compounds with radioisotope iodium-131 as a cancer therapy agent and determine the optimum condition of marking until it obtains high marking efficiency to achieve therapeutic goals. In this experiment, curcumin compounds have been successfully labelled by Na-1311 through the chloramine-Tmethod. The marking efficiency and radiochemical purity of compounds is determined by paper electrophoresis methods and thin-layer chromatography using ITLC-SG plates as a silent phase and NaCl 0.9% as the motion phase. Test results were detected with TLC-radio scanner (AR 2000) with optimal marking conditions obtained at the use of 0.1mg/100µL curcumin, 100 uci Na-1311, 0.625mg/40µL chloramin-T and sodium metabisulfite, as well as an incubation time of 90 minutes at 25-30°C. The average percent purity obtained was 90.62% ±0.77. The results of lipophilicity test showed a decrease in lipophilic properties of curcumin, while from the results of electrophoresis showed negatively charged compound 1311-curcumin. Based on the test results, it can be concluded that the marked compound has met the requirements of radiochemical purity of radiopharmaceutical preparations according to the Indonesian Pharmacopeia where the minimum requirement of radiochemical purity set is >90%.
