Abstract

Biosurfaktan merupakan alternatif surfaktan sintesis yang disintesis secara ekstraseluler oleh mikroorganisme yang memiliki aktivitas sebagai penurun tegangan permukaan. Biosurfaktan dari mikroorganisme mempunyai sifat fisika kimia yang stabil, tidak mencemari lingkungan, sangat mudah terurai, dapat stabil pada temperatur tinggi, kadar asam dan garam tinggi, bahan baku dapat diperbaharui, lebih murah dan memiliki toksisitas rendah. Tujuan penelitian ini adalah mendapatkan bakteri yang berpotensi sebagai biosurfaktan dan punya aktivitas antimikroba. Pada penelitian ini terhadap bakteri Bacillus cereus dan Brevundimonas terrae dilakukan uji hemolisis dan oil spreading test. Kemudian dilakukan uji aktivitas biosurfaktan menggunakan metode emulsifikasi. Terhadap supernatan bakteri dilakukan uji kromatografi lapis tipis dan antimikroba. Hasil yang didapat menunjukkan persentase emulsifikasi bakteri Bacillus cereus dan Brevundimonas terrae masing-masing pada hari ketiga yaitu 51,72% dan 48,28%. Dapat disimpulkan kedua bakteri ini berpotensi sebagai biosurfaktan dan memiliki aktivitas antimikroba. Hasil kromatografi lapis tipis menandakan biosurfaktan termasuk golongan lipopeptida yang ditandai spot berwarna merah setelah disemprot reagen ninhidrin.;---Biosurfactant is an alternative surfactant synthesis that is synthesized extracellularly by microorganisms which have a surface tension-reducing activity. Biosurfactants from microorganisms have stable physical chemical properties, do not pollute the environment, are highly biodegradable, stabilize at high temperatures, have high levels of acid and salt, renewable raw materials, cheaper and have low toxicity. The purpose of this study was to obtain bacteria that have the potential as biosurfactants and antimicrobial activity. In this study Bacillus cereus and Brevundimonas terrae bacteria were carried out by hemolysis and oil spreading test. Then biosurfactant test activity was tested by using the emulsification method. Bacterial supernatants were carried out by thin layer chromatography and antimicrobial activity. The results obtained were the emulsification percentage of Bacillus cereus and Brevundimonas terrae bacteria on the third day at 51.72% and 48.28% respectively. In conclusion this bacterium has potential as biosurfactant and has antimicrobial activity. The results of thin layer chromatography indicate that biosurfactants are a class of lipopeptides which are marked with red spots after being sprayed with ninhydrin reagent.
