Abstract

Kurkumin (1,7-bis (4’ hidroksi-3 metoksifenil)-1,6 heptadien, 3,5-dion) dikenal sebagai hasil isolasi bahan alam yang mempunyai aktivitas biologis diantaranya sebagai antioksidan, antivirus, antiinflamasi, antimikroba, dan antikanker. Kelarutan kurkumin dalam air yang rendah dapat menyebabkan kelarutan dan absorbsi pada saluran cerna menjadi rendah, sehingga dibuat kurkumin larut air dengan cara reaksi penggaraman atau disebut dengan soluble kurkumin. Untuk mengetahui meningkatkannya bioavailabilitas dan juga efek farmakologinya maka salah satu upaya yang dapat dilakukan adalah menguji keamanan melalui uji toksisitas akut oral. Pengujian uji toksisitas dilakukan dengan menggunakan 4 ekor mencit Swiss Webster betina. Kelompok tersebut terdiri dari kontrol normal diberi Aquadest, kelompok uji terbatas (Limit test) diberikan isolat soluble kurkumin dengan dosis >5000 mg/kgBB. Pengujian dilakukan selama 14 hari. Menggunakan metode OECD 425 Up and Down Procedure. Data yang diperoleh dianalisis dengan menggunakan AOT425StatPgm untuk menentukan estimasi nilai LD50. Berdasarkan hasil pengujian uji toksisitas akut oral isolat soluble kurkumin nilai LD50 yaitu >5000 mg/kgBB termasuk kriteria penggolongan sediaan praktis tidak toksik. Secara makroskopik dan mikroskopik tidak teramati gejala-gejala toksisitas pada organ hati, lambung dan ginjal.;---Curcumin (1,7-bis (4 'hydroxy-3 methoxyphenyl) -1.6 heptadien, 3,5-Dion) is known as a result of the isolation of natural substances that have biological activities that contain antioxidants, antiviral, anti-inflammatory, antimicrobial, and anticancer properties. The low solubility of curcumin in water can cause low solubility and absorption in the gastrointestinal tract so that air-soluble curcumin is made using a salting reaction or called soluble curcumin. To find out about increasing bioavailability and pharmacological effects, one of the efforts that can be done is to use safety through oral toxicity testing. Toxicity testing was performed using 4 female Swiss Webster mice. The group consisted of normal controls given Aquadest, a limited test group (limit test) given dissolved curcumin with a dose >5000 mg/kgBB. Testing is carried out for 14 days. Using the OECD 425 Up and Down Procedure. The data obtained were analyzed using AOT425Statggm to determine the estimated LD50 value. Based on the results of acute toxicity testing of oral isolates that can dissolve curcumin LD50 values >5000 mg/kgBB body weight including criteria for the classification of practically non-toxic preparations. Macroscopically and microscopically there are no symptoms of toxicity to the liver, stomach and kidney.
