Abstract

Etil p-metoksisinamat merupakan salah satu metabolit sekunder dan komponen utama yang dikandung oleh rimpang kencur (Kaempferia galanga L.) dalam jumlah yang relatif besar. Telah dilakukan pengujian toksisitas akut isolat etil p-metoksisinamatuntuk mengetahui LD50 terhadap hewan percobaan mencit Swiss Webster betina dengan menggunakan metode terbaru standar OECD 425 Up and Down Proceduredengan aplikasi software AOT425StatPgm Versi.1.0. Hewan dibagi menjadi dua kelompok yaitu kelompok normal yang diberikan Na-CMC 0,5% dan kelompok bahan uji yang diberikan isolat etil p-metoksisinamat secara oral. Pengamatan tanda-tanda toksisitas dilakukan dengan jangka pendek (48 jam) dan jangka panjang (14 hari). Hasil penelitian menunjukkan bahwa nilai LD50 dari isolat etil p-metoksisinamat adalah >5000 mg/KgBB, termasuk ke dalam klasifikasi praktis tidak toksik.Pada pemeriksaan histopatologi, berdasarkan nilai rata-rata skoring menunjukkan hasil bahwa pemberian isolat etil p-metoksisisnamat tidak berpengaruh terhadap organ hati, lambung dan ginjal pada mencit.;---Ethyl p-methoxycinnamate is one of the secondary metabolites and the main component contained in kencur (Kaempferia galanga L.) in a relatively large amount. An acute toxicity test of ethyl p-methoxycinnamate isolate has been carried out to determine LD50 in experimental animals of female Swiss Webster mice used the latest OECD 425 Up and Down Procedure standard with the application of AOT425StatPgm Version.1.0 software. Animals were divided into two groups: the normal group given 0.5% Na-CMC and the group of test material given ethyl p-methoxycinnamate isolate orally. Observation of signs of toxicity is done in the short term (48 hours) and long term (14 days). The results showed that the LD50 value of ethyl p-methoxycinnamate isolate was> 5000 mg / KgBW, included in the practically non-toxic classification. On histopathological examination, based on the average score, the results showed that the administration of ethyl p-methoxycinnamate isolate had no effect on the liver, stomach and kidney in mice.
