Abstract
Piperin merupakan senyawa aktif utama yang terkandung dalam Piper nigrum L. dan memiliki aktivitas antiinflamasi. Namun, piperin termasuk dalam Biopharmaceutics Classification System (BCS) kelas II dengan karakteristik permeabilitas tinggi dan kelarutan rendah sehingga proses disolusi dan absorpsinya menjadi terbatas. Salah satu pendekatan untuk mengatasi keterbatasan tersebut adalah pembentukan kompleks inklusi menggunakan natrium karagenan. Penelitian ini bertujuan mengevaluasi keberhasilan pembentukan kompleks inklusi piperin– natrium karagenan serta aktivitas antiinflamasinya pada model edema telinga tikus putih (Rattus norvegicus) yang diinduksi xylene. Karakterisasi kompleks inklusi dilakukan melalui pengujian kadar air, kadar piperin, kelarutan, dan Fourier Transform Infrared (FTIR). Aktivitas antiinflamasi dievaluasi berdasarkan perubahan ketebalan dan lebar edema telinga, nilai Area Under the Curve (AUC), serta persentase inhibisi edema. Data dianalisis menggunakan Mixed ANOVA (Repeated Measures ANOVA) yang dilanjutkan dengan uji Pairwise Comparisons. Hasil penelitian menunjukkan bahwa pembentukan kompleks inklusi meningkatkan kelarutan piperin sebesar 2,07 kali dibandingkan piperin murni. Karakterisasi FTIR menunjukkan adanya interaksi antara piperin dan natrium karagenan tanpa pembentukan gugus fungsi baru. Analisis statistik menunjukkan bahwa kompleks inklusi menunjukkan respons antiinflamasi yang lebih besar dibandingkan piperin murni, ditandai dengan penurunan edema yang lebih besar, nilai AUC yang lebih rendah, serta persentase inhibisi edema yang lebih tinggi (p < 0,05). Dengan demikian, pembentukan kompleks inklusi piperin–natrium karagenan terbukti meningkatkan sifat fisikokimia piperin dan aktivitas antiinflamasinya. ------- Piperine is the major bioactive compound found in Piper nigrum L. and possesses anti-inflammatory activity. However, piperine is classified as a Biopharmaceutics Classification System (BCS) Class II compound, characterized by high permeability and low aqueous solubility, which limits its dissolution and absorption. One approach to overcome this limitation is the formation of an inclusion complex using sodium carrageenan. This study aimed to evaluate the successful formation of the piperine–sodium carrageenan inclusion complex and its anti-inflammatory activity in a xylene-induced ear edema model in white rats (Rattus norvegicus). The inclusion complex was characterized by determining moisture content, piperine content, solubility, and Fourier Transform Infrared (FTIR) spectroscopy. Antiinflammatory activity was evaluated based on changes in ear edema thickness and width, Area Under the Curve (AUC), and percentage of edema inhibition. The data were analyzed using Mixed ANOVA (Repeated Measures ANOVA) followed by Pairwise Comparisons. The results showed that the inclusion complex increased the solubility of piperine by 2.07-fold compared with pure piperine. FTIR analysis indicated the interaction between piperine and sodium carrageenan without the formation of new functional groups. Statistical analysis demonstrated that the inclusion complex showed a greater anti-inflammatory response compared with pure piperine, as indicated by greater edema reduction, lower AUC values, and higher percentages of edema inhibition (p < 0.05). In conclusion, the piperine– sodium carrageenan inclusion complex improved the physicochemical properties of piperine, particularly its solubility, and enhanced its anti-inflammatory activity.
