Abstract
Daun cengkeh (Syzygium aromaticum L.) diketahui mengandung senyawa fenolik dan flavonoid yang berpotensi sebagai antioksidan. Umur daun menjadi salah satu faktor yang dapat mempengaruhi kandungan senyawa sehingga berpengaruh terhadap aktivitasnya. Penelitian ini bertujuan membandingkan kadar total fenol, kadar total flavonoid, serta aktivitas antioksidan ekstrak daun cengkeh muda dan tua serta menganalisis korelasinya. Kadar total fenol ditentukan menggunakan metode Folin-Ciocalteu, kadar total flavonoid menggunakan metode AlCl₃, sedangkan aktivitas antioksidan menggunakan metode DPPH. Data dianalisis menggunakan Independent Sample t-test dan korelasi Pearson. Hasil penelitian menunjukkan kadar total fenol ekstrak daun muda dan tua secara berturut-turut sebesar 643,779 mg GAE/g dan 705,282 mg GAE/g, kadar total flavonoid sebesar 13,822 mg QE/g dan 15,836 mg QE/g, serta nilai IC₅₀ sebesar 12,102 ppm dan 7,725 ppm. Hasil menunjukkan terdapat perbedaan yang signifikan (Sig. (2-tailed) < 0,05) pada ketiga parameter yang diuji antara kedua ekstrak dan adanya korelasi yang sangat kuat terhadap nilai IC50. Kesimpulan penelitian ekstrak daun tua memiliki kandungan fenol, flavonoid, dan aktivitas antioksidan yang lebih tinggi dibandingkan daun muda. ------- Clove leaves (Syzygium aromaticum L.) are known to contain phenolic compounds and flavonoids that have antioxidant potential. The age of the leaves is one factor that can influence the content of these compounds, thereby affecting their activity. This study aimed to compare the total phenolic content, total flavonoid content, and antioxidant activity of extracts from young and old clove leaves and to determine their correlation. Total phenolic content was determined using the Folin-Ciocalteu method, total flavonoid content using the AlCl₃ method, and antioxidant activity using the DPPH method. Data were analyzed using the independent samples t-test and Pearson’s correlation. The results showed that the total phenolic content of the young and old leaf extracts was 643,779 mg GAE/g and 705,282 mg GAE/g, respectively; the total flavonoid content was 13,822 mg QE/g and 15,836 mg QE/g, and IC₅₀ values of 12,102 ppm and 7,725 ppm, respectively. The results indicate a significant difference (Sig. (2-tailed)< 0.05) in all three tested parameters between the two extracts and a very strong correlation with the IC₅₀ values. The study concludes that the extract from older leaves has higher levels of phenolic compounds, flavonoids, and antioxidant activity compared to that from younger leaves.
