Abstract
------ Catechin isolates from Uncaria gambir Roxb. have the potential to be developed as active ingredients in facial toner formulations due to their high antioxidant activity. The physical characteristics of the toner are influenced by the composition of the excipients, particularly glycerin and propylene glycol as humectants. This study aimed to determine the effect of variations in the composition of glycerin and propylene glycol on the viscosity and pH of the catechin isolate facial toner and to identify the optimal formulation using the Simplex Lattice Design (SLD) method. The study was conducted using five formulas designed with Design Expert® version 13, varying the compositions of glycerin and propylene glycol. Evaluations included organoleptic testing, homogeneity, pH, viscosity, formula optimization, model validation, a cycling test, and determination of catechin content. The results showed that all five formulations exhibited good organoleptic properties, were homogeneous, had a pH of 5.28-5.53, and a viscosity of 1.15-1.40 cP, thereby meeting the quality requirements for a facial toner. Variations in glycerin and propylene glycol significantly affected viscosity (p = 0.0035) but did not significantly affect pH. The optimal formulation was obtained with a composition of 11% glycerin and 6% propylene glycol and demonstrated good physical stability during the cycling test. The conclusion of this study is that variations in glycerin and propylene glycol affect viscosity but do not affect pH, and that the optimal formulation of 11% glycerin and 6% propylene glycol meets quality requirements and exhibits good physical stability.
