Abstract

Kurkumin merupakan senyawa aktif dari rimpang kunyit (Curcuma longa L.) yang memiliki banyak aktivitas farmakologis, tetapi penggunaannya masih terbatas karena kelarutan dan bioavailabilitasnya rendah. Selain itu, data terkait penetrasi perkutan kurkumin masih terbatas, sedangkan kurkumin memiliki aktivitas antiiflamasi dan antioksidan yang potensial untuk sediaan topikal. Penelitian ini bertujuan memperoleh profil penetrasi perkutan kurkumin dengan penambahan piperin sebagai bioenhancer dan menentukan rasio yang menghasilkan penetrasi tertinggi. Uji penetrasi dilakukan secara in vitro menggunakan Sel Difusi Franz dengan membran kulit mencit, media reseptor dapar fosfat pH 7,4 dengan suhu 37°C dan analisis kadar menggunakan spektrofotometri UV-Vis pada 420 nm. Sampel diuji pada rasio kurkumin:piperin 1:0, 1:1, 1:5, dan 1:10 selama 120 menit. Hasil menunjukkan nilai Q/A pada menit ke-120 pada perbandingan 1:0, 1:1, 1:5, dan 1:10 berturut-turut sebesar 3,1941 ± 0,1555; 7,7042 ± 0,2532; 14,5354 ± 0,1155; dan 35,9835 ± 0,1643 µg/cm². Nilai steady-state flux (Jss) pada kurkumin pada perbandingan 1:0, 1:1, 1:5, dan 1:10 berturut-turut sebesar 0,027446 ± 0,000255; 0,035326 ± 0,002876; 0,061234 ± 0,000072; dan 0,190272 ± 0,001013 µg/cm²/menit, dengan enhancement ratio tertinggi sebesar 6,93 pada rasio 1:10. Berdasarkan penelitian ini, dapat disimpulkan bahwa penambahan piperin menunjukkan kecenderungan meningkatkan penetrasi kurkumin, dan rasio 1:10 memberikan hasil tertinggi dalam rentang rasio yang diuji. ------ Curcumin is an active compound derived from turmeric rhizome (Curcuma longa L.) with anti-inflammatory and antioxidant activities; however, its use is limited by poor solubility and low bioavailability. Data related to the percutaneous penetration of curcumin is still limited, while curcumin has potential antiinflammatory and antioxidant activities for topical preparations. This study characterized the percutaneous penetration profile of curcumin with piperine and identified the ratio producing the highest penetration. Testing was conducted in vitro using Franz diffusion cells with ex vivo mouse skin, phosphate buffer pH 7.4 as receptor medium, a temperature of 37°C, and UV-Vis spectrophotometric analysis at 420 nm. Curcumin:piperine ratios of 1:0, 1:1, 1:5, and 1:10 were tested for 120 minutes. The cumulative amounts penetrated per unit area at 120 minutes were 3.1941 ± 0.1555, 7.7042 ± 0.2532, 14.5354 ± 0.1155, and 35.9835 ± 0.1643 µg/cm², respectively. The steady-state flux values were 0.027446 ± 0.000255, 0.035326 ± 0.002876, 0.061234 ± 0.000072, and 0.190272 ± 0.001013 µg/cm²/min, respectively, with the highest enhancement ratio of 6.93 at 1:10. Descriptively, increasing piperine ratios increased cumulative penetration, average flux, permeability coefficient, and percentage diffused compared with the control. Piperine tended to enhance curcumin penetration, with the 1:10 ratio producing the highest result within the tested range.
