Abstract
Plasma kuda hiperimun merupakan bahan baku penting dalam produksi Serum Anti Bisa Ular (SABU) karena mengandung imunoglobulin G (IgG) sebagai sumber antibodi yang dapat diproses menjadi fragmen F(ab’)₂. Penelitian ini bertujuan mengevaluasi pengaruh suhu penyimpanan terhadap profil fisikokimia plasma dan stabilitas protein IgG. Sampel plasma disimpan pada suhu 2–8 °C, 20–25 °C, dan 37 °C, kemudian dianalisis melalui organoleptis, pH, konduktivitas, OD280, OD600, Dynamic Light Scattering (DLS), SDS-PAGE non-reduksi, densitometri, dan RP-HPLC. Data OD280, OD600, dan DLS dianalisis menggunakan Two Way ANOVA, sedangkan SDS-PAGE/densitometri dan RP-HPLC dianalisis secara semikuantitatif dan deskriptif. Hasil menunjukkan bahwa organoleptis, pH, konduktivitas, dan OD600 relatif stabil selama penyimpanan, sedangkan OD280, DLS, SDS-PAGE/densitometri, dan RP-HPLC menunjukkan perubahan profil protein, partikel, dan kromatogram. Perubahan paling nyata terjadi pada suhu 37 °C. Berdasarkan parameter yang diuji, suhu 2–8 °C merupakan kondisi penyimpanan yang paling mendukung, terutama ditunjukkan oleh OD600 yang relatif stabil serta pita protein pada wilayah IgG utuh 150 kDa ± 10 kDa yang masih terdeteksi hingga minggu ke-8. Hasil ini dapat menjadi dasar awal pertimbangan batas waktu penyimpanan sementara plasma sebelum produksi SABU, tetapi belum menetapkan umur simpan definitif. ------ Hyperimmune horse plasma is an important raw material in the production of Snake Antivenom Serum (SABU) because it contains immunoglobulin G (IgG), which can be processed into F(ab’)₂ fragments. This study aimed to evaluate the effect of storage temperature on the physicochemical profile of plasma and IgG protein stability. Plasma samples were stored at 2–8 °C, 20–25 °C, and 37 °C, then analyzed using organoleptic observation, pH, conductivity, OD280, OD600, Dynamic Light Scattering (DLS), non-reducing SDS-PAGE, densitometry, and RPHPLC. OD280, OD600, and DLS data were analyzed using Two Way ANOVA, while SDS-PAGE/densitometry and RP-HPLC were analyzed semiquantitatively and descriptively. The results showed that organoleptic properties, pH, conductivity, and OD600 were relatively stable during storage, whereas OD280, DLS, SDS-PAGE/densitometry, and RP-HPLC showed changes in protein, particle, and chromatogram profiles. The most noticeable changes occurred at 37 °C. Based on the tested parameters, 2–8 °C was the most supportive storage condition, mainly indicated by relatively stable OD600 values and the protein band in the intact IgG region of 150 kDa ± 10 kDa that was still detected until week 8. These results can serve as an initial basis for temporary plasma storage before SABU production, but have not established a definitive shelf life.
