Abstract
Penelitian ini bertujuan untuk menentukan kadar fenol dan flavonoid total dari ekstrak daun sukun kuning jatuh (Artocarpus altilis (Parkinson) Fosberg), bunga rosella (Hibiscus sabdariffa L.), serta kombinasinya dengan perbandingan 1:1, 1:2, dan 2:1. Ekstraksi dilakukan menggunakan metode dekokta hingga diperoleh ekstrak kental, kemudian dilakukan analisis menggunakan spektrofotometri UVVis. Kadar fenol total ditetapkan dengan metode Folin–Ciocalteu menggunakan asam galat sebagai standar pada panjang gelombang 765 nm, sedangkan kadar flavonoid total ditetapkan dengan metode kolorimetri menggunakan kuersetin sebagai standar pada panjang gelombang 415 nm. Hasil penelitian menunjukkan bahwa kombinasi ekstrak 1:2 (daun sukun:rosella) memiliki kadar fenol total tertinggi sebesar 49,925 mg GAE/g ekstrak dan kadar flavonoid total tertinggi sebesar 47,714 mg QE/g ekstrak. Hasil ini menunjukkan adanya efek sinergis antara kedua ekstrak, di mana peningkatan proporsi bunga rosella mampu meningkatkan kadar senyawa fenolik dan flavonoid yang berpotensi memberikan aktivitas antioksidan lebih optimal. ------- This study aimed to determine the total phenolic and flavonoid contents of extracts from yellow fallen breadfruit leaves (Artocarpus altilis (Parkinson) Fosberg), roselle flowers (Hibiscus sabdariffa L.), and their combinations in ratios of 1:1, 1:2, and 2:1. Extraction was carried out using the decoction method until a concentrated extract was obtained, followed by analysis using UV-Vis spectrophotometry. The total phenolic content was determined by the Folin–Ciocalteu method using gallic acid as a standard at a wavelength of 765 nm, while the total flavonoid content was determined by the colorimetric method using quercetin as a standard at a wavelength of 415 nm. The results showed that the 1:2 (breadfruit leaves:roselle) combination had the highest total phenolic content of 49.925 mg GAE/g extract and the highest total flavonoid content of 47.714 mg QE/g extract. These findings indicate a synergistic effect between both extracts, where an increased proportion of roselle enhances the levels of phenolic and flavonoid compounds, suggesting greater antioxidant potential..
