Abstract

Radikal bebas merupakan molekul reaktif yang dapat merusak komponen seluler dan berkontribusi pada timbulnya penyakit degeneratif. Antioksidan berperan penting dalam menangkal kerusakan tersebut, sehingga eksplorasi sumber alami menjadi fokus penelitian. Ampas ekstrak lada putih (Piper nigrum L.), yang umumnya dianggap limbah, diketahui masih mengandung metabolit sekunder seperti alkaloid dan saponin yang berpotensi sebagai antioksidan. Penelitian ini bertujuan menentukan aktivitas antioksidan ampas ekstrak lada putih menggunakan metode Ferric Reducing Antioxidant Power (FRAP) berbasis microplate reader, serta membandingkannya dengan vitamin C sebagai kontrol positif. Skrining fitokimia menunjukkan ampas masih mengandung alkaloid dan saponin, meskipun flavonoid, terpenoid, dan polifenol tidak terdeteksi. Hasil uji FRAP memperlihatkan peningkatan aktivitas antioksidan seiring kenaikan konsentrasi. Ampas ekstrak lada putih memiliki nilai IC₅₀ sebesar 11,24 µg/mL dengan FRAP value maksimum 13.031 µmol Fe²⁺/g, sedangkan vitamin C menunjukkan IC₅₀ sebesar 29,03 µg/mL dengan FRAP value 32.113 µmol Fe²⁺/g. Meskipun kapasitas reduktif vitamin C lebih tinggi, ampas ekstrak lada putih menunjukkan efisiensi dosis yang lebih baik. Temuan ini menegaskan bahwa ampas ekstrak lada putih berpotensi sebagai sumber alternatif antioksidan alami sekaligus mendukung pemanfaatan limbah berbasis zero waste untuk aplikasi farmasi dan kesehatan. ------ Free radicals are highly reactive molecules capable of damaging cellular components and contributing to the development of degenerative diseases. Antioxidants play a crucial role in counteracting these effects, making the exploration of natural sources essential. White pepper (Piper nigrum L.) extraction residue, commonly regarded as waste, is known to still contain secondary metabolites such as alkaloids and saponins with antioxidant potential. This study aimed to determine the antioxidant activity of white pepper residue using the Ferric Reducing Antioxidant Power (FRAP) method with a microplate reader, and to compare it with vitamin C as a positive control. Phytochemical screening revealed that the residue retained alkaloids and saponins, while flavonoids, terpenoids, and polyphenols were undetectable. FRAP assay results showed an increase in antioxidant activity with increasing concentrations. The residue exhibited an IC₅₀ value of 11.24 µg/mL with a maximum FRAP value of 13,031 µmol Fe²⁺/g, whereas vitamin C showed an IC₅₀ of 29.03 µg/mL with a FRAP value of 32,113 µmol Fe²⁺/g. Although vitamin C demonstrated higher reducing capacity, the residue displayed better dose efficiency. These findings indicate that white pepper extraction residue has potential as an alternative natural antioxidant while supporting zero waste utilization for pharmaceutical and health applications.
