Abstract

Kanker prostat merupakan salah satu penyebab utama kematian pada pria di seluruh dunia. Brazilin, senyawa aktif dari kayu secang (Caesalpinia sappan L.), diketahui memiliki potensi sebagai agen antikanker melalui mekanisme induksi apoptosis. Penelitian ini bertujuan untuk mengevaluasi pengaruh brazilin terhadap mekanisme kematian sel pada sel kanker prostat DU-145 secara in vitro dengan metode flow cytometry. Sel DU-145 dikelompokkan menjadi tiga kelompok perlakuan: kontrol (DMSO), kontrol positif (docetaxel), dan perlakuan brazilin. Analisis viabilitas dan apoptosis dilakukan menggunakan pewarnaan Annexin VFITC/PI yang dibaca dengan flow cytometry. Hasil penelitian menunjukkan bahwa kelompok kontrol mempertahankan viabilitas tinggi 90,23% dengan apoptosis rendah 9,36%. Perlakuan docetaxel menurunkan viabilitas hingga 77,80% dan meningkatkan apoptosis menjadi 21,57%. Sementara itu, brazilin lebih efektif dengan penurunan viabilitas menjadi 67,03% dan peningkatan apoptosis hingga 29,51%, terutama pada tahap apoptosis lanjut. Temuan ini menunjukkan bahwa brazilin berpotensi sebagai kandidat agen antikanker alami yang dapat menjadi alternatif atau komplementer dalam terapi kanker prostat. ------- Prostate cancer is one of the leading causes of death among men worldwide. Brazilin, an active compound isolated from Caesalpinia sappan L., has been recognized for its potential as an anticancer agent by inducing apoptosis. This study aimed to evaluate the effect of brazilin on cell death mechanisms in DU-145 prostate cancer cells in vitro using flow cytometry. DU-145 cells were divided into three treatment groups: control (DMSO), positive control (docetaxel), and brazilin treatment. Cell viability and apoptosis were analyzed using Annexin V-FITC/PI staining and quantified by flow cytometry. The results demonstrated that the control group maintained high cell viability (90.23%) with low apoptosis (9.36%). Treatment with docetaxel reduced cell viability to 77.80% and increased apoptosis to 21.57%. Interestingly, brazilin showed a stronger effect by decreasing cell viability to 67.03% and elevating apoptosis to 29.51%, predominantly in the late apoptotic phase. These findings suggest that brazilin possesses significant potential as a natural anticancer candidate and may serve as an alternative or complementary therapeutic option for prostate cancer
