Abstract

Senyawa antioksidan memiliki peran penting dalam melindungi sel-sel tubuh dari kerusakan akibat radikal bebas. Salah satu tanaman yang berpotensi sebagai antioksidan alami adalah rimpang bangle (Zingiber cassumunar Roxb.). Penelitian ini bertujuan untuk menganalisis pengaruh perbedaan wilayah tumbuh terhadap aktivitas antioksidan serta karakterisasi minyak atsiri rimpang bangle (Zingiber cassumunar Roxb.). Sampel yang digunakan berupa rimpang bangle dari Kecamatan Langkaplancar, Pangandaran, Jawa Barat; Kecamatan Wedi, Klaten, Jawa Tengah; dan Kecamatan Gemarang, Madiun, Jawa Timur. Ekstraksi minyak atsiri dilakukan dengan metode destilasi uap air. Pengujian aktivitas antioksidan dilakukan menggunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl). Uji aktivitas antioksidan dilakukan menggunakan microplate reader pada panjang gelombang 517 nm dan Trolox sebagai kontrol positif. Hasil penelitian menunjukkan bahwa terdapat perbedaan nilai IC₅₀ antar sampel, dengan nilai IC₅₀ berturut-turut sebesar 92,30 ppm (Jawa Barat), 62,73 ppm (Jawa Tengah), dan 305,64 ppm (Jawa Timur). Berdasarkan analisis statistik (p > 0,05), perbedaan tersebut tidak signifikan secara statistik, namun secara deskriptif sampel dari Jawa Tengah memiliki nilai IC₅₀ paling rendah, yang menunjukkan kecenderungan aktivitas antioksidan lebih kuat dibandingkan sampel dari Jawa Barat dan Jawa Timur. Selain itu, hasil karakterisasi minyak atsiri rimpang bangle menunjukkan adanya variasi antarwilayah tumbuh. Perbedaan ini diduga dipengaruhi oleh faktor lingkungan tumbuh, seperti kondisi tanah, intensitas cahaya, dan kelembaban udara di masing-masing lokasi. ----- Antioxidant compounds play an important role in protecting body cells from damage caused by free radicals. One plant with potential as a natural antioxidant is bangle rhizome (Zingiber cassumunar Roxb.). This study aims to analyze the effect of growing region differences on antioxidant activity and characterize the essential oil of bangle rhizome (Zingiber cassumunar Roxb.). The samples used were bangle rhizomes from Langkaplancar District, Pangandaran, West Java; Wedi District, Klaten, Central Java; and Gemarang District, Madiun, East Java. Essential oil extraction was performed using the water steam distillation method. Antioxidant activity testing was performed using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. Antioxidant activity testing was performed using a microplate reader at a wavelength of 517 nm and Trolox as a positive control. The results showed that there were differences in IC₅₀ values between samples, with IC₅₀ values of 92.30 ppm (West Java), 62.73 ppm (Central Java), and 305.64 ppm (East Java), respectively. Based on statistical analysis (p > 0.05), these differences were not statistically significant, but descriptively, the sample from Central Java had the lowest IC₅₀ value, indicating a tendency for stronger antioxidant activity compared to samples from West Java and East Java. In addition, the results of the characterization of bangle rhizome essential oil showed variations between growing regions. These differences are thought to be influenced by environmental factors, such as soil conditions, light intensity, and air humidity in each location.
