Abstract
Sambiloto (Andrographis paniculata), memiliki berbagai fungsi farmakologis seperti anti-inflamasi dan antioksidan. Untuk meningkatkan efektivitas dan kemurniannya, pemurnian crude andrografolid menjadi sangat penting. Penelitian ini bertujuan mengevaluasi efektivitas metode penjerapan menggunakan Karbon Aktif dan Silika Gel 60 Tipe G sebagai alternatif pemurnian crude andrografolid yang dihasilkan Sekolah Tinggi Farmasi Indonesia (STFI). Metode yang digunakan meliputi Kromatografi Cair Vakum (KCV) dengan fase diam Silika Gel G dan eluen n-heksan:Etil asetat, memanfaatkan karbon aktif sebagai adsorben untuk menghilangkan kontaminan. Fraksi hasil KCV kemudian dianalisis kemurniannya menggunakan Kromatografi Lapis Tipis (KLT), Uji Titik Leleh, Spektrofotometri UV-Vis, dan Spektroskopi Inframerah Transformasi Fourier (FTIR). Hasil KLT menggunakan fase gerak Kloroform:Metanol (7:3) menunjukkan nilai Rf 0,84, yang berbeda dari standar andrografolid (Rf ±0,45), mengindikasikan pemisahan belum optimal karena eluen terlalu polar. Namun, uji titik leleh menunjukkan rentang sempit (231,4°C), yang mengindikasikan tingkat kemurnian yang tinggi. Analisis UV menunjukkan puncak absorbansi maksimum pada 228 nm, sesuai karakteristik andrografolid. Spektrum FTIR mengkonfirmasi adanya gugus fungsi karakteristik seperti hidroksil (O-H) pada 3388 cm⁻¹ dan karbonil (C=O) pada 1746 cm⁻¹, mendukung identifikasi andrografolid.Secara keseluruhan, metode penjerapan, khususnya KCV yang dikombinasikan dengan Karbon Aktif, berhasil mengisolasi senyawa dengan tingkat kemurnian fisik (titik leleh) dan karakteristik kimia (UV-Vis dan FTIR) yang mendekati andrografolid murni, meskipun optimalisasi fase gerak KLT lebih lanjut diperlukan. ------- (Andrographis paniculata) possesses various pharmacological functions such as anti-inflammatory and antioxidant properties. To enhance its effectiveness and purity, the purification of crude andrographolide is crucial.This research aimed to evaluate the effectiveness of an adsorption method utilizing Activated Carbon and Silica Gel 60 Type G as an alternative for purifying crude andrographolide produced by the School of Pharmacy Indonesia (Sekolah Tinggi Farmasi Indonesia, STFI).Methodology The purification method involved Vacuum Liquid Chromatography (VLC) using Silica Gel G as the stationary phase and Nhexane:Ethyl acetate as the eluent, with Activated Carbon utilized as an adsorbent to remove contaminants. The purity of the resulting fractions from VLC was then analyzed using Thin Layer Chromatography (TLC), Melting Point Test, UV-Vis Spectrophotometry, and Fourier Transform Infrared (FTIR) Spectroscopy.Results and AnalysisTLC Analysis: Results from TLC using the mobile phase Chloroform:Methanol (7:3) showed an Rf value of 0.84. This value differed from the andrographolide standard (Rf ±0.45), indicating that the separation was not yet optimal because the eluent was too polar.Melting Point Test: Despite the sub-optimal TLC result, the melting point test displayed a narrow range (231.4°C), which suggests a high level of physical purity in the isolated compound. UV-Vis Analysis: The UV-Vis analysis showed a maximum absorbance peak at 228 nm, consistent with the characteristic of andrographolide.FTIR Spectroscopy: The FTIR spectrum confirmed the presence of characteristic functional groups such as hydroxyl (O−H) at 3388 cm−1 and carbonyl (C=O) at 1746 cm−1, further supporting the identification of andrographolide.
