Abstract
Kanker prostat merupakan salah satu penyebab utama kematian pada pria, sehingga dibutuhkan alternatif terapi berbasis bahan alam yang lebih aman. Alfa mangostin, senyawa bioaktif dari kulit buah manggis (Garcinia mangostana L.), memiliki aktivitas antikanker melalui induksi apoptosis. Penelitian ini bertujuan untuk mengetahui pengaruh alfa mangostin terhadap mekanisme kematian sel pada sel kanker prostat DU-145 menggunakan metode Flow Cytometry. Sel DU-145 diberi perlakuan dengan alfa mangostin pada konsentrasi IC50 (14,17 µg/mL), kontrol DMSO (negatif), dan kontrol docetaxel (positif) 21,27 µg/mL. Pewarnaan Annexin V-FITC dan Propidium Iodide (PI) digunakan untuk analisis mekanisme kematian sel. Hasil menunjukkan bahwa alfa mangostin meningkatkan apoptosis total menjadi 13,90%, lebih tinggi dibandingkan kontrol DMSO (negatif) 9,36%, tetapi lebih rendah dibandingkan docetaxel 21,57%. Nekrosis yang diinduksi alfa mangostin relatif rendah 1,07%, ini menunjukkan bahwa mekanisme kematian sel terjadi secara spesifik melalui jalur apoptosis, bukan akibat toksisitas non-spesifik. Dengan demikian, alfa mangostin berpotensi sebagai agen antikanker alami, meskipun efektivitasnya masih di bawah docetaxel. ------ Prostate cancer is a major cause of mortality in men, requiring safer natural based therapeutic alternatives. Alpha mangostin, a bioactive compound from the pericarp of Garcinia mangostana L., is known for its anticancer activity through apoptosis induction. This study aimed to evaluate the effect of mangostin on cell death mechanisms in DU-145 prostate cancer cells using Flow Cytometry. DU-145 cells were treated with Alpha mangostin at IC50 concentration (14.17 µg/mL), DMSO control (negative), and docetaxel control (positive) 21.27 µg/mL. Cell death mechanism were analyzed using Annexin V-FITC and Propidium Iodide (PI) staining. Results showed that mangostin increased total apoptosis to 13.90%, higher than the DMSO control (negative) 9.36%, but lower than docetaxel 21.57%. Necrosis remained low 1,07%, indicating a specific apoptotic pathway rather than non-specific cytotoxicity. In conclusion, alpha mangostin demonstrated potential as a natural anticancer agent by inducing apoptosis in DU-145 prostate cancer cells, although its efficacy was still lower than docetaxel.
