Abstract

Kulit buah manggis (Garcinia mangostana L.) diketahui mengandung metabolit bioaktif dengan aktivitas antioksidan dan antibakteri, namun kajian mengenai bakteri endofit yang hidup di dalam jaringan kulit buah masih terbatas. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri endofit dari kulit buah manggis serta mengetahui karakteristik fenotipiknya. Isolasi dilakukan melalui metode sterilisasi permukaan dan penanaman pada media TSA, dilanjutkan pemurnian isolat dan uji identifikasi makroskopis, mikroskopis, fisiologis, serta biokimia (katalase, koagulase, pigmentasi, toleransi NaCl, pertumbuhan pada suhu 15°C dan 45 °C). Hasil isolasi diperoleh empat isolat (EM1, EM2, EM3, EM4) dengan variasi morfologi koloni, seluruhnya menunjukkan Gram positif berbentuk kokus, katalase positif, koagulase positif, serta toleran terhadap NaCl tinggi. Hasil analisis dengan aplikasi ABIS menunjukkan bahwa keempat isolat memiliki kemiripan 99% dengan Staphylococcus aureus subsp. aureus. ------ Mangosteen peel (Garcinia mangostana L.) is known to contain bioactive metabolites with antioxidant and antibacterial properties; however, studies on endophytic bacteria inhabiting its peel tissue remain limited. This study aimed to isolate and identify endophytic bacteria from mangosteen peel and to determine their phenotypic characteristics. Isolation was carried out through surface sterilization and cultivation on TSA medium, followed by purification and macroscopic, microscopic, physiological, and biochemical characterization (catalase, coagulase, pigmentation, NaCl tolerance, and growth at 15°C and 45°C). Four isolates (EM1, EM2, EM3, EM4) were obtained with distinct colony morphologies. All isolates were Gram-positive cocci, catalase positive, coagulase positive, and tolerant to high NaCl concentrations. Identification using ABIS software confirmed that all isolates showed 99% similarity to Staphylococcus aureus subsp. aureus.
