Abstract
Penyalahgunaan narkotika, khususnya kokain (COC) masih menjadi masalah global yang berdampak serius terhadap kesehatan masyarakat. Deteksi kokain yang cepat, akurat, dan spesifik sangat dibutuhkan untuk mendukung penanggulangan penyalahgunaan narkotika. Salah satu metode yang banyak digunakan adalah Lateral Flow Assay (LFA) yaitu deteksi sederhana, cepat, dan mudah diaplikasikan. Salah satu pendekatan yang menjanjikan adalah penggunaan antibodi rekayasa berupa Single Chain Fragment Variable (scFv). Penelitian ini bertujuan untuk menentukan serta mengevaluasi afinitas dan spesifisitas fragmen antibodi scFv M82G2 terhadap kokain dibandingkan dengan narkotika jenis lainnya yang sering disalahgunakan (seperti metamfetamin, morfin, benzodiazepin, amfetamin, dan THC) menggunakan metode in silico. Hasil penambatan molekul pada kokain memiliki energi ikatan bebas (ΔG) – 6,62 kkal/mol, interaksi scFv M82G2-kokain memiliki afinitas cukup baik. Perbandingan dengan ligan lain menunjukkan bahwa metamfetamin memiliki nilai (ΔG) –4,75 kkal/mol dan benzodiazepin nilai (ΔG) –5,50 kkal/mol menunjukkan afinitas lebih rendah, sedangkan morfin nilai (ΔG) -6.27 kkal/mol dan amfetamin nilai (ΔG) -5.74 kkal/mol menunjukkan afinitas sedang. Sementara itu, THC memiliki nilai (ΔG) lebih rendah –8,00 kkal/mol akibat dominasi interaksi hidrofobik. Hal ini menunjukkan bahwa meskipun pada penelitian sebelumnya kokain telah berikatan dengan scFv M82G2, tetapi antibodi ini masih berpotensi berinteraksi dengan senyawa non-target sehingga dapat menimbulkan reaktivitas silang dalam aplikasi diagnostik. ------ Drug abuse, particularly cocaine (COC), remains a global problem that has a serious impact on public health. Rapid, accurate, and specific detection of cocaine is urgently needed to support efforts to combat drug abuse. One widely used method is the Lateral Flow Assay (LFA), which is simple, fast, and easy to apply. One promising approach is the use of engineered antibodies in the form of Single Chain Fragment Variable (scFv). This study aims to determine and evaluate the affinity and specificity of the scFv M82G2 antibody fragment to cocaine compared to other commonly abused drugs (such as methamphetamine, morphine, benzodiazepine, amphetamine, and THC) using an in silico method. The results of molecular binding to cocaine had a free binding energy (ΔG) of –6.62 kcal/mol, indicating that the scFv M82G2-cocaine interaction had fairly good affinity. Comparison with other ligands shows that methamphetamine has a value of (ΔG) –4.75 kcal/mol and benzodiazepine has a value of (ΔG) –5.50 kcal/mol, indicating lower affinity, while morphine has a value of (ΔG) -6.27 kcal/mol and amphetamine has a value of (ΔG) -5.74 kcal/mol, indicating moderate affinity. Meanwhile, THC has a lower value (ΔG) of –8.00 kcal/mol due to the dominance of hydrophobic interactions. This indicates that although cocaine has been shown to bind to scFv M82G2 in previous studies, this antibody still has the potential to interact with non-target compounds, which could cause cross-reactivity in diagnostic applications.
