Abstract
Daun sukun (Artocarpus altilis (Parkinson) Fosberg) dan bunga rosella (Hibiscus sabdariffa L.) memiliki potensi sebagai terapi antikolesterol melalui kandungan flavonoid dan antosianin. Namun, informasi mengenai keamanan kombinasi kedua ekstrak ini masih terbatas. Penelitian ini bertujuan untuk mengetahui toksisitas akut kombinasi ekstrak daun sukun dan bunga rosella dengan rasio 2:1 pada tikus Wistar betina. Uji dilakukan dengan metode Acute Toxic Class (OECD 423) pada dosis 2000 mg/kgBB dan 5000 mg/kgBB secara oral, kemudian diamati selama 14 hari terhadap gejala klinis, mortalitas, perubahan berat badan, indeks organ, serta gambaran histopatologi hati, ginjal, jantung, paru-paru, dan limpa. Hasil penelitian menunjukkan bahwa pemberian kombinasi ekstrak hingga dosis 5000 mg/kgBB tidak menimbulkan mortalitas maupun gejala toksik. Pemeriksaan histopatologi memperlihatkan degenerasi parenkimatosa ringan pada hati, edema ringan pada ginjal, hemoragi dan degenerasi ringan pada jantung, inflamasi ringan pada paruparu, serta penurunan diameter pulpa alba limpa yang tidak signifikan terhadap efek toksik. Nilai LD₅₀ diperkirakan lebih dari 5000 mg/kgBB, sehingga kombinasi ekstrak dikategorikan tidak toksik secara akut. Penelitian ini menunjukkan bahwa kombinasi ekstrak daun sukun dan bunga rosela relatif aman pada pemberian akut, meskipun uji lanjutan tetap diperlukan untuk memastikan keamanan jangka panjangnya. ------- Breadfruit leaves (Artocarpus altilis) and roselle (Hibiscus sabdariffa L.) possess potential anticholesterol activity due to their flavonoid and anthocyanin contents. However, information regarding the safety of their combined extract remains limited. This study aimed to evaluate the acute toxicity of a 2:1 combination of breadfruit leaf and roselle flower extracts in female Wistar rats. The test was conducted using the Acute Toxic Class method (OECD 423) at oral doses of 2000 and 5000 mg/kg body weight, with 14 days of observation for clinical signs, mortality, body weight changes, organ index, and histopathological alterations in the liver, kidney, heart, lung, and spleen. The results showed that administration up to 5000 mg/kg caused no mortality or toxic clinical signs. Histopathological examination revealed mild parenchymatous degeneration in the liver, mild edema in the kidney, mild hemorrhage and degeneration in the heart, mild inflammation in the lung, and a slight decrease in spleen white pulp diameter, which were not significant to toxic effects. The estimated LD₅₀ value was greater than 5000 mg/kg, indicating that the combination is classified as nontoxic under acute exposure. This study suggests that the combination of Artocarpus altilis and Hibiscus sabdariffa L. extracts is relatively safe under acute administration; however, further studies with larger samples, biochemical analyses, and subchronic to chronic testing are required to confirm its long-term safety.
