Abstract

Kulit manggis memiliki berbagai senyawa bioaktif terutama pada bagian kulit buahnya, seperti xanthone, flavonoid dan tannin yang berpotensi sebagai antioksidan. Tujuan penelitian ini untuk mengisolasi dan mengidentifikasi jamur endofit dari kulit buah manggis (Garcinia mangostana L.) dengan pendekatan morfologis dan molekuler. Proses isolasi dilakukan melalui sterilisasi permukaan jaringan kulit buah manggis, kemudian ditanam pada media Malt Extract Agar (MEA). Identifikasi makroskopis menunjukkan koloni jamur berwarna putih menyerupai kapas dengan perbedaan pola pertumbuhan. Hasil pengamatan mikroskopis memperlihatkan hifa bersepta bercabang serta struktur reproduksi aseksual berupa konidia dan fialid. Analisis molekuler dilakukan melalui amplifikasi gen ITS dengan hasil fragmen DNA ±550 bp. Hasil BLAST menunjukkan query coverage 100%, percent identity 100%, E-value 0.0, dan max score 1026, yang mengidentifikasi isolat sebagai Neofusicoccum parvum (filum Ascomycota). Pohon filogenetik mengidentifikasi kekerabatan isolat jamur endofit GMLF04 dari kulit buah manggis (Garcinia mangostana L.). Hasil analisis menggunakan metode Neighbor-Joining menunjukkan bahwa GMLF04 berkerabat dekat dengan Neofusicoccum parvum, tergabung dalam satu klad bersama beberapa strain lainnya dengan nilai bootstrap 62–84%. ------ The rind of mangosteen (Garcinia mangostana L.) contains various bioactive compounds, particularly xanthones, flavonoids, and tannins, which have potential as antioxidants. This study aimed to isolate and identify endophytic fungi from the mangosteen rind using morphological and molecular approaches. The isolation process was carried out through surface sterilization of the rind tissue, followed by cultivation on Malt Extract Agar (MEA) medium. Macroscopic identification showed white, cotton-like fungal colonies with varying growth patterns. Microscopic observations revealed branched, septate hyphae and asexual reproductive structures in the form of conidia and phialides. Molecular analysis was performed by amplifying the ITS (Internal Transcribed Spacer) gene, resulting in a DNA fragment of approximately 550 bp. BLAST analysis showed 100% query coverage, 100% percent identity, an E-value of 0.0, and a maximum score of 1026, identifying the isolate as Neofusicoccum parvum (phylum Ascomycota). Phylogenetic tree identified the relationship of endophytic fungal isolate GMLF04 from mangosteen (Garcinia mangostana L.) peel. The analysis results using the Neighbor-Joining method showed that GMLF04 was closely related to Neofusicoccum parvum, grouped in one clade with several other strains with a bootstrap value of 62–84%.
