Abstract

Ikan gabus (Channa striata) dikenal sebagai sumber protein hewani dengan kandungan albumin yang tinggi. Albumin merupakan protein utama yang berperan penting dalam pembentukan jaringan baru dan mempercepat regenerasi sel yang rusak. Beberapa penelitian sebelumnya menyatakan bahwa kandungan albumin pada ikan gabus memiliki potensi dalam mendukung proses penyembuhan luka. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak etanol freeze dry ikan gabus (Channa striata) terhadap percepatan penyembuhan luka pada hewan uji. Penelitian menggunakan mencit jantan yang dibagi ke dalam lima kelompok perlakuan, yaitu 4% ekstrak etanol freeze dry ikan gabus, 8% ekstrak etanol freeze dry ikan gabus, kontrol positif (bioskin gel), kontrol negatif (basis Na-CMC 3%), dan kontrol normal (tanpa perlakuan). Luka dibuat pada punggung mencit dengan metode punch biopsy, berikan perlakuan sesuai kelompok uji. Pengamatan dilakukan dengan mengukur diameter luka serta menganalisis gambaran hispatologi pada hari ke – 7. Hasil pengukuran menunjukkan bahwa kelompok dengan gel ekstrak 8% memiliki persentase penyembuhan luka paling tinggi (45,0 ± 10,1%) dan diameter luka paling kecil. Uji Kruskal-Wallis menunjukkan tidak ada perbedaan signifikan antar kelompok perlakuan (p=0,168), mengindikasikan bahwa semua perlakuan, termasuk ekstrak memiliki potensi penyembuhan. Analisis hispatologi memperlihatkan bahwa gel 8% merupakan konsentrasi paling optimal dengan skor epitelisasi (3) dan angiogenesis (4) tertinggi dibanding gel 4%. Efektivitas ini didukung oleh kemampuan gel 8% mengendalikan inflamasi secara efisien, ditandai dengan penurunan infiltrasi neutrofil dan makrofag. Ekstrak etanol freeze dry ikan gabus terbukti efektif mempercepat proses penyembuhan luka dengan konsentrasi 8% merupakan dosis yang paling optimal. ----- Snakehead fish (Channa striata) is known as an animal protein source with a high content of albumin. Albumin is a major protein that plays an important role in the formation of new tissues and accelerates the regeneration of damaged cells. Several previous studies have reported that the albumin content in snakehead fish has potential in supporting the wound healing process. This study aimed to determine the effectiveness of ethanol freeze-dried extract of Channa striata in accelerating wound healing in experimental animals. The study used male mice, which were divided into five treatment groups: 4% ethanol freeze-dried extract of Channa striata, 8% ethanol freeze-dried extract of Channa striata, positive control (bioskin gel), negative control (3% Na-CMC base), and normal control (without treatment). Wounds were created on the dorsal skin of the mice using the punch biopsy method, treatment according to each group. Observations were carried out by measuring wound diameter and analyzing histopathological features on day 7. The results showed that the 8% extract gel group achieved the highest wound healing percentage (45.0 ± 10.1%) and the smallest wound diameter. The Kruskal-Wallis test showed no significant differences among the treatment groups (p = 0.168), indicating that all treatments, including the extract, had healing potential. Histopathological analysis demonstrated that the 8% gel was the most optimal concentration, with the highest epithelialization score (3) and angiogenesis score (3) compared to 4% gel. This effectiveness was supported by the ability of the 8% gel to efficiently control inflammation, as indicated by decreased infiltration of neutrophils and macrophages. Ethanol freeze-dried extract of Channa striata was proven effective in accelerating the wound healing process, with 8% concentration being the most optimal dose.
